The sequence incorporates alpha-aminoisobutyric acid at position 8, replacing the alanine found in the natural hormone. This substitution blocks the primary DPP-4 recognition site and contributes most of the enzymatic stability. Albumin binding further protects the peptide and reduces the frequency of administration required to maintain active plasma levels. Because the fatty acid chain increases lipophilicity, the compound is formulated as a solution rather than a simple aqueous buffer. Researchers describe the design as an incremental optimization of earlier GLP-1 analogs rather than a wholly new scaffold.
Reported molecular weight is approximately 4113.6 daltons for the free base, and the peptide is supplied as a lyophilized powder or in buffered liquid form depending on the intended use. It is freely soluble in water when formulated with appropriate excipients, though the unconjugated peptide shows limited stability at neutral pH over long periods. Analytical characterization typically relies on reversed-phase high-performance liquid chromatography and mass spectrometry. Purity specifications for research-grade material commonly exceed ninety-five percent by area. Isotopic and impurity profiles differ between suppliers.
Two principal therapeutic variants exist under separate regulatory filings, one indicated for glycemic control in type 2 diabetes and one for chronic weight management. Both use the same active molecule; differences lie in formulation strength, titration schedule, and labeling. Regulatory agencies in the United States and European Union approved injectable forms in 2017 and 2018 respectively. An oral tablet formulation received approval later, using a carrier molecule to enhance absorption across the gastric epithelium. Labeling differs by jurisdiction and by indication.
Stability studies examine how temperature, pH, and moisture influence degradation rates over time. In aqueous solution, hydrolysis and deamidation accelerate as pH moves away from mildly acidic conditions. Light exposure and residual metal ions can also trigger oxidation of susceptible residues. Accelerated aging at elevated temperature is used to estimate shelf life, though extrapolation to room temperature carries uncertainty because individual degradation pathways do not always scale predictably.
Lyophilized semaglutide powder is typically held at minus twenty degrees Celsius for long-term storage. At that temperature, solid-state degradation reactions proceed slowly and the peptide remains intact for extended periods. Repeated freeze-thaw cycles are best avoided because they promote aggregation and can shift the proportion of monomeric peptide present. Working aliquots are often prepared so that each portion is thawed only once, and desiccant is placed inside the vial to limit moisture uptake.
Identity and purity are usually assessed by reverse-phase high-performance liquid chromatography coupled to mass spectrometry. Retention time and observed mass are compared against a reference standard run under identical conditions. Impurity profiles reveal deamidation products, oxidized methionine variants, and truncated fragments that arise during synthesis or storage. Peptide mapping through enzymatic digestion confirms the primary sequence, while amino acid analysis offers an independent check on overall composition.

The semaglutide sequence is a 31-residue analogue of human GLP-1, altered at three positions relative to the parent hormone. Aminoisobutyric acid replaces alanine at position 8, arginine replaces lysine at position 34, and a lipophilic diacid is attached to lysine 26 through a short linker. These features are reported consistently in the structural literature. The position 8 substitution blocks recognition by dipeptidyl peptidase-4, while the attached chain drives strong, reversible association with a carrier protein in blood.
Receptor activation raises intracellular cyclic AMP through Gs coupling, which promotes glucose-dependent insulin release and suppresses glucagon secretion when blood glucose is elevated. Effects outside the pancreas include slower gastric emptying and altered appetite signalling in the hypothalamus and hindbrain. The relative contribution of each tissue to overall metabolic outcomes remains an area of active investigation. Central mechanisms in particular are inferred mainly from animal models and indirect human measures rather than direct observation.
Serum protein binding dominates the pharmacokinetic profile. The attached chain associates strongly with albumin, shielding the peptide from enzymatic attack and slowing filtration by the kidney. This interaction extends the circulation half-life to roughly one week in humans, which supports weekly administration intervals. An oral version pairs the peptide with an absorption enhancer that transiently alters gastric epithelium, permitting limited uptake; bioavailability by that route is substantially lower than by injection.
Semaglutide is a synthetic peptide analogue of glucagon-like peptide-1, a gut hormone released by intestinal L cells after food intake. The natural hormone acts on pancreatic and central receptors but is degraded within minutes by dipeptidyl peptidase-4 and other peptidases. Semaglutide belongs to the class of long-acting GLP-1 receptor agonists, a group distinguished by structural changes that slow breakdown and extend circulation time. Its development followed earlier short-acting analogues and reflects a general strategy in peptide drug design: preserve receptor activity while blocking proteolytic clearance.
Three structural changes define the molecule. At position 8 an alpha-aminoisobutyric acid residue replaces alanine, which blocks dipeptidyl peptidase-4 cleavage. At position 34 arginine replaces lysine, and at position 26 a lysine carries a C18 fatty diacid attached through a short linker. The fatty chain binds serum albumin, and this albumin association reduces renal filtration and enzymatic attack. The unchanged backbone retains the receptor contacts that produce signalling. The free base has the formula C187H291N45O59 and a molecular weight near 4114 daltons.
Semaglutide is a synthetic peptide analogue of glucagon-like peptide-1, a gut hormone released after nutrient intake. The molecule contains 31 amino acid residues and differs from the native sequence at several positions. A non-natural residue at position eight resists the enzyme that normally truncates the hormone, while a lysine-linked fatty diacid side chain promotes binding to serum albumin. These two modifications extend the circulating half-life from minutes to roughly one week. The peptide is produced by solid-phase synthesis followed by selective acylation, and its identity and purity are confirmed by spectrometric and chromatographic techniques.
The primary target is the GLP-1 receptor, a class B G protein-coupled receptor expressed on pancreatic beta cells, in the gut, and in several brain regions. Receptor activation raises intracellular cyclic AMP, which potentiates glucose-dependent insulin secretion and lowers glucagon release when blood glucose is elevated. Signalling in the hypothalamus and brainstem is associated with reduced appetite and slower gastric emptying. Because the insulinotropic effect depends on prevailing glucose levels, the hypoglycaemic risk of the peptide alone is described as low in most study settings. The relative contribution of peripheral and central actions remains an active research question.
Large randomised trials in adults with type 2 diabetes and in adults with obesity have reported reductions in body weight and improvements in several cardiovascular risk markers. One outcome trial found a lower incidence of major adverse cardiovascular events in participants with diabetes and established cardiovascular disease. Gastrointestinal effects such as nausea and vomiting are the most frequently reported adverse events and often diminish over time. Changes in lean body mass during weight loss are an area of ongoing investigation. Effects in adolescents and in pregnancy are less well characterised, and current labelling advises against use during pregnancy.
Somatotrophs, which secrete growth hormone (a peptide hormone) Lactotrophs, which secrete prolactin (a peptide hormone) When using standard staining techniques, they cannot be distinguished from each other (though they can be distinguished from basophils and chromophobes), and are therefore identified simply as "acidophils".

Males perform a sequence of five behavioral patterns to court females. First, males orient themselves while playing a courtship song by horizontally extending and vibrating their wings. Soon after, the male positions himself at the rear of the female's abdomen in a low posture to tap and lick the female genitalia. Finally, the male curls his abdomen and attempts copulation. Females can reject males by moving away, kicking, and extruding their ovipositor. Copulation lasts around 15–20 minutes, during which males transfer a few hundred, very long (1.76 mm) sperm cells in seminal fluid to the female. Females store the sperm in a tubular receptacle and in two mushroom-shaped spermathecae; sperm from multiple matings compete for fertilization. A last male precedence is believed to exist; the last male to mate with a female sires about 80% of her offspring. This precedence was found to occur through both displacement and incapacitation. The displacement is attributed to sperm handling by the female fly as multiple matings are conducted and is most significant during the first 1–2 days after copulation. Displacement from the seminal receptacle is more significant than displacement from the spermathecae. Incapacitation of first male sperm by second male sperm becomes significant 2–7 days after copulation. The seminal fluid of the second male is believed to be responsible for this incapacitation mechanism (without removal of first male sperm) which takes effect before fertilization occurs.
Linus Carl Pauling was born on February 28, 1901, in Portland, Oregon, the firstborn child of Herman Henry William Pauling (1876–1910) and Lucy Isabelle "Belle" Darling (1881–1926). He was named "Linus Carl", in honor of Lucy's father, Linus, and Herman's father, Carl. His ancestry included German and English-Scottish. In 1902, after his sister Pauline was born, Pauling's parents decided to move out of Portland to find more affordable and spacious living quarters than their one-room apartment. Lucy stayed with her husband's parents in Lake Oswego until Herman brought the family to Salem, where he worked briefly as a traveling salesman for the Skidmore Drug Company. Within a year of Lucile's birth in 1904, Herman Pauling moved his family to Lake Oswego, Oregon where he opened his own drugstore. He moved his family to Condon, Oregon, in 1905. By 1906, Herman Pauling was suffering from recurrent abdominal pain. He died of a perforated ulcer on June 11, 1910, leaving Lucy to care for Linus, Lucile and Pauline. Pauling attributes his interest in becoming a chemist to being amazed by experiments conducted by a friend, Lloyd A. Jeffress, who had a small chemistry lab kit. He later wrote: "I was simply entranced by chemical phenomena, by the reactions in which substances, often with strikingly different properties, appear; and I hoped to learn more and more about this aspect of the world." In high school, Pauling conducted chemistry experiments by scavenging equipment and material from an abandoned steel plant.

With an area of 22,402,200 square kilometres (8,649,500 sq mi), the Soviet Union was the world's largest country, a status that is retained by the Russian Federation. Covering a sixth of Earth's land surface, its size was comparable to that of North America. Two other successor states are also very large — Kazakhstan ranks among the top 10 countries by land area, and Ukraine is the largest country entirely in Europe. The European portion accounted for a quarter of the country's area and was the cultural and economic centre. The eastern part in Asia extended to the Pacific Ocean to the east and Afghanistan to the south, and, except some areas in Central Asia, was much less populous. It spanned over 10,000 kilometres (6,200 mi) east to west across 11 time zones, and over 7,200 kilometres (4,500 mi) north to south. It had five climate zones: tundra, taiga, steppes, desert and mountains. The USSR, like Russia, had the world's longest border, measuring over 60,000 kilometres (37,000 mi), or 1+1⁄2 circumferences of Earth. Two-thirds of it was a coastline. The country bordered Afghanistan, the People's Republic of China, Czechoslovakia, Finland, Hungary, Iran, Mongolia, North Korea, Norway, Poland, Romania, and Turkey from 1945 to 1991. The Bering Strait separated the USSR from the United States, while the La Pérouse Strait separated it from Japan. The country's highest mountain was Communism Peak (now Ismoil Somoni Peak) in Tajikistan, at 7,495 metres (24,590 ft).
=== 1918 influenza pandemic === The influenza pandemic of 1918 placed a sever strain on Oregon’s hospitals and public health infrastructure. At the time, the viral cause of influenza had not yet been identified, and many physicians believed that the high mortality rate resulted from secondary bacterial infections. In response, the Oregon State Board of Health supplied Lawrence’s laboratory with an infected tissue sample obtained from a navy yard in Bremerton, Washington. Lawrence isolated hemolytic streptococcus from patient samples and developed a vaccine targeting this bacterium, which was associated with secondary pneumonia in influenza patients. Although described at the time as an influenza vaccine, the preparation was designed to combat bacterial infections that frequently followed influenza rather than the virus itself. The vaccines produced in her laboratory were distributed to physicians throughout Oregon as part of broader public health efforts. Her work received national recognition, and she was later honored by President Woodrow Wilson for her contributions.
== Alternative barcodes to DNA == DNA barcodes can have limitations. Many standard chemical reactions can degrade DNA and thus compromise the chemical barcodes, necessitating changing chemical reaction conditions that could alter the binding ability of the small molecule to its target. Additionally, the DNA tag is typically over 50 times larger than the molecule itself, potentially restricting the binding ability of each library member and sometimes interacting with the target itself, creating false hits or obscuring potentially otherwise strong binders. This is especially problematic when the target has nucleic acid binding sites, like transcription factors or RNA-binding proteins. For this reason, a multitude of barcode alternatives have been developed in efforts to mitigate these issues such as abiotic peptides, peptide nucleic acids, and even barcode free self-encoded libraries.
== Society and culture == Paroxetine was approved for medical use in the United States in 1992 and initially sold by GlaxoSmithKline. It is available as a generic medication. In 2022, it was the 92nd most commonly prescribed medication in the United States, with more than 7 million prescriptions. It is on the World Health Organization's List of Essential Medicines. GlaxoSmithKline has paid substantial fines, paid settlements in class-action lawsuits, and become the subject of several highly critical books about its marketing of paroxetine, in particular, the off-label marketing of paroxetine for children, the suppression of negative research results relating to its use in children, and allegations that it failed to warn consumers of substantial withdrawal effects associated with the use of the drug.
=== Legal status === Pegulicianine was approved for medical use in the United States in April 2024. The US Food and Drug Administration granted the application for pegulicianine fast track and priority review designations.

The rapid expansion of AI infrastructure has significant environmental implications. The energy demands of data centres are projected to grow substantially, potentially conflicting with the UK's net-zero targets and straining grid capacity. Additionally, the water consumption required for cooling data centres has raised sustainability concerns. Beyond domestic impacts, the global AI supply chain relies heavily on the extraction of critical minerals (such as cobalt and lithium) in the Global South, often associated with environmental degradation and human rights abuses. The development of AI models also depends on low-wage "ghost workers" in developing nations for data annotation and content moderation, prompting calls for mandatory human rights due diligence laws. While economic forecasts suggest AI could significantly boost UK GDP and productivity, realising these gains faces a "productivity paradox". Despite experimental evidence of productivity improvements, economy-wide data does not yet reflect an AI-driven boost, largely due to low adoption rates of bespoke systems among traditional businesses and the concentration of economic value among a small percentage of organisations. Furthermore, economists warn of "algorithmic rents", where dominant tech platforms extract wealth, potentially eroding the UK tax base and exacerbating market concentration. The UK's Digital Services Tax (DST) attempts to address this but remains controversial with the US government.
Diagnostic Measurement Tools – Clinical psychologists are able to diagnose psychological disorders and related disorders found in the DSM-5 and ICD-10. Many assessment tests have been developed to complement the clinicians clinical observation and other assessment activities. Some of these include the SCID-IV, the MINI, as well as some specific to certain psychological disorders such as the CAPS-5 for trauma, the ASEBA, and the K-SADS for affective and Schizophrenia in children. Clinical observation – Clinical psychologists are also trained to gather data by observing behavior. The clinical interview is a vital part of the assessment, even when using other formalized tools, which can employ either a structured or unstructured format. Such assessment looks at certain areas, such as general appearance and behavior, mood and affects, perception, comprehension, orientation, insight, memory, and content of the communication. One psychiatric example of a formal interview is the mental status examination, which is often used in psychiatry as a screening tool for treatment or further testing.
== History == In literature from ancient Rome there are complaints of foul air, while in 1273 the inhabitants of London were discussing the prohibition of coal burning to improve air quality. However, the measurement and analysis of aerosols only became established in the second half of the 19th century. In 1847 Henri Becquerel presented the first concept of particles in the air in his condensation nuclei experiment and his ideas were confirmed in later experiments by Coulier in 1875. These ideas were expanded on between 1880 and 1890 by meteorologist John Aitken who demonstrated the fundamental role of dust particles in the formation of clouds and fogs. Aitken's method for aerosol analysis consisted of counting and sizing particles mounted on a slide, using a microscope. The composition of the particles was determined by their refractive index. In the 1920s aerosol measurements, using Aitken's simple microscopic method, became more common place because the negative health effects of industrial aerosols and dust were starting to be recognized by health organizations. Technological and instrumentation advancements, including improved filters, led to improvement in the aerosol measurement methods in the 1960s. The introduction of polycarbonate filters, called nucleopore filters, enhanced the collection, storage, and transportation of samples without disturbing the physical and chemical state of the particles. On-line aerosol measurements methods took a little longer than off-line to be developed and perfected. It wasn't till 1973 with W.D.
==== Pharmacokinetics of pyridine derivatives ==== Axitinib has short half-life, ranging from 2.5 to 6.1 hours, and therefore steady state should be reached in 2–3 days after the first dose. Peak plasma concentration is reached in 2.5 to 4.1 hours. In vitro protein binding is over 99%. Axitinib is primarily metabolized in the liver by CYP3A4/5. 30-60% of the drug is excreted in faeces, and about 23% in urine. Regorafenib is a pyridine derivative, but also a urea derivative and has therefore been covered in that section.

It is a modified version of the natural hormone, with three amino acid changes and a fatty acid side chain added. These edits extend its half-life from minutes to about one week. The core receptor activity is retained.
Both deliver the same active peptide and act on the same receptor. The tablet includes an absorption enhancer because peptides are poorly taken up intact from the gut. Bioavailability of the oral route is substantially lower, so the two are not dose-equivalent.
No, it is entirely synthetic and does not occur in nature. Native GLP-1 is produced in the gut and pancreas, but the analog is manufactured by chemical synthesis or recombinant methods. Traces of the analog are not expected in people who never received it.
Manufacturers commonly state multi-year stability when the powder is kept dry and frozen. Actual shelf life depends on residual moisture, vial sealing, and storage temperature. A re-test by chromatography is the only way to confirm remaining purity.
