=== Immune system === Inflammation has been suggested to have a role in the pathogenesis of fibromyalgia. People with fibromyalgia tend to have higher levels of inflammatory cytokines IL-6, and IL-8. There are also increased levels of the pro-inflammatory cytokines IL-1 receptor antagonist. Increased levels of pro-inflammatory cytokines may increase sensitivity to pain, and contribute to mood problems. Anti-inflammatory interleukins such as IL-10 have also been associated with fibromyalgia. Neurogenic inflammation has been proposed as a contributing factor to fibromyalgia. A repeated observation shows that autoimmunity triggers, such as traumas and infections, are among the most frequent events preceding the onset of fibromyalgia. == Causes == SJS is thought to arise from a disorder of the immune system. The immune reaction can be triggered by drugs or infections. Genetic factors are associated with a predisposition to SJS such as individuals expressing the human leukocyte antigen (i.e. HLA) serotypes HLA-B15:02 or HLA-A31:01 The cause of SJS is unknown in one-quarter to one-half of cases. SJS, SJS/TEN, and TEN are considered a single disease with common causes and mechanisms. Individuals with variations in their efficiency to absorb, distribute to tissues, metabolize, or excrete a drug are predisposed to develop SJS.
In either case, if the concentration of the irreversible antagonist is high enough, the number of unbound receptors remaining for agonist binding may be so low that even high concentrations of the agonist do not produce the maximum biological response.
