Quantification and purity assessment rely on separation methods coupled to optical or mass detection. Reversed-phase high-performance liquid chromatography resolves the intact peptide from related impurities and is the standard assay technique. Size-exclusion chromatography measures aggregates, while ion-exchange chromatography separates charge variants produced by deamidation. Mass spectrometry confirms identity and detects mass shifts of a few daltons. In biological matrices, liquid chromatography with tandem mass spectrometry is often used because immunoassays can cross-react with endogenous GLP-1 or with circulating fragments.
As a peptide, semaglutide is sensitive to conditions that break amide bonds or modify side chains. Deamidation of asparagine and glutamine residues, oxidation of methionine and tryptophan, and non-covalent aggregation are the main degradation routes described in published stability work. Rate depends strongly on pH, buffer species, ionic strength, temperature and exposure to light. Formulators therefore choose a defined solution pH and often add excipients such as phosphate buffer, propylene glycol and phenol, each of which plays a separate role in pH control, tonicity or preservation.
Storage guidance for the finished injectable product distinguishes the unused state from the in-use state. Before first use, pens are kept refrigerated between 2 and 8 degrees Celsius, protected from light, and never frozen, since freezing can disrupt the peptide or the device. After first use, label instructions in several markets permit storage at room temperature up to about 30 degrees Celsius for a limited number of days. Solid research-grade material is normally held at or below minus 20 degrees Celsius, often with desiccant, and allowed to equilibrate before opening.
Clinical studies of semaglutide generally measure glycated hemoglobin, fasting plasma glucose, body weight, and composite cardiovascular endpoints. The SUSTAIN program enrolled adults with type 2 diabetes, while the STEP program focused on obesity without diabetes. Administration follows a stepwise escalation schedule designed to limit gastrointestinal effects during the first weeks. Reported outcomes include mean percentage weight change, the proportion of participants reaching defined weight-loss thresholds, and rates of nausea, vomiting, and diarrhea. Long-term data on durability after treatment stops are still limited and remain a topic of ongoing research.
Semaglutide is a synthetic peptide analog of human glucagon-like peptide-1, developed by Novo Nordisk and first approved in 2017 for type 2 diabetes. It belongs to the incretin mimetic class, a group of agents that reproduce the glucose-dependent actions of endogenous GLP-1. The molecule was engineered to resist degradation by dipeptidyl peptidase-4 and to bind serum albumin, extending its half-life from minutes to roughly one week. Approval for chronic weight management followed in 2021, based on large cardiovascular and obesity outcome trials.

Stability studies focus on deamidation of asparagine and glutamine residues, oxidation of methionine, and aggregation into higher-order species. The fatty acid side chain adds susceptibility to oxidative change and can promote self-association at high concentration. Lyophilised material is comparatively robust when kept cold and dry, while aqueous solutions require refrigeration and protection from light. Forced degradation experiments under heat, acid, base, and peroxide conditions establish the specificity of each analytical method. Which degradation route dominates under real storage conditions depends on the formulation and stays formulation-specific.
Handling guidance for research quantities calls for single-use aliquots, an inert atmosphere where practical, and avoidance of repeated freeze-thaw cycles that accelerate aggregation. Certificates of analysis typically report purity by peak area, water content, counter-ion identity, and residual solvent levels. In the scientific literature the compound is usually described by its full amino acid sequence, its registry number, or its structural class rather than by any proprietary label. Reporting standards vary between journals, and reviewers increasingly request raw chromatograms alongside tabulated purity figures. Whether current purity thresholds are adequate for every experimental context is debated.
Reversed-phase high-performance liquid chromatography with ultraviolet detection is the dominant approach for peptide purity assessment, usually paired with mass spectrometry to confirm molecular mass and sequence. Peptide mapping by enzymatic digestion and tandem mass spectrometry locates modifications such as deamidation and oxidation. Quantitation in plasma matrices can be performed by LC-MS/MS after solid-phase extraction. Method validation follows general guidance on accuracy, precision, linearity, and limits of detection. Comparability of results between laboratories, when no shared reference standard is available, remains an open question.

=== Pharmacokinetics === LC–MS is widely used in the field of bioanalysis and is specially involved in pharmacokinetic studies of pharmaceuticals. Pharmacokinetic studies are needed to determine how quickly a drug will be cleared from the body organs and the hepatic (liver) blood flow. MS analyzers are useful in these studies because of their shorter analysis time, and higher sensitivity and specificity compared to UV detectors commonly attached to HPLC systems. One major advantage is the use of tandem MS–MS, where the detector may be programmed to select certain ions to fragment. The measured quantity is the sum of molecule fragments chosen by the operator. As long as there are no interferences or ion suppression in LC–MS, the LC separation can be done quite swiftly.
Mimoun "Rocky" Kaabouni, who was nicknamed "The Moroccan Escobar", was born in Ben Taïeb, a village in the province of Nador. He grew up in Hoensbroek, in the Netherlands, where he and his brother owned a coffeeshop named Happy Days. While often visiting Amsterdam, he eventually started meeting big names within the Moroccan mafia with whom he maintained regular contact. Mimoun left the Netherlands for southern Spain and Romania, where he founded his own criminal organisation made up of Lithuanians and Colombians. Mimoun, who was already bringing in tons of cocaine through the ports of Antwerp, Rotterdam, Amsterdam, and Algeciras, wanted to create a new route in Romania through the port of Constanța. He is currently serving a 19-year prison sentence in Romania for a record seizure of 2.5 tons of cocaine (the biggest seizure of cocaine in the country's history). Mimoun was arrested in 2016 when he tried to leave Spain through the Málaga Airport after the seizure in Romania, which was worth €600 million. It was one of the largest drug seizures in Eastern European history.
== Synthesis == Endorphin precursors are primarily produced in the pituitary gland. All three types of endorphins are fragments of the precursor protein proopiomelanocortin (POMC). At the trans-Golgi network, POMC binds to a membrane-bound protein, carboxypeptidase E (CPE). CPE facilitates POMC transport into immature budding vesicles. In mammals, proprotein convertase 1 (PC1) cleaves POMC into adrenocorticotropin (ACTH) and beta-lipotropin (β-LPH). β-LPH, a pituitary hormone with little opioid activity, is then continually fragmented into different peptides, including α-endorphin, β-endorphin, and γ-endorphin. Proprotein convertase 2 (PC2) is responsible for cleaving β-LPH into β-endorphin and γ-lipotropin. Formation of α-endorphin and γ-endorphin results from proteolytic cleavage of β-endorphin.
The following mechanism shows a proposed scheme for a metal-dependent "pita-bread" enzyme with residue numbering corresponding to those found in methionine aminopeptidase from E. coli. As shown in Intermediate I of the figure, three potential acidic amino acid residues interact with the N-terminus of the substrate in a fashion that is yet to be determined. The carbonyl and amide groups of the scissile peptide bond interact with the first metal ion, M1, in addition to His178 and His79, respectively. M1 and Glu204 activate a water molecule to prepare it nucleophilic attack at the carbonyl carbon of the scissile peptide bond. Then, the tetrahedral intermediate (Intermediate II) becomes stabilized from interactions with M1 and His178. Lastly, Glu204 donates a proton to the amine of the leaving peptide (P1'). This leads to the breakdown of the intermediate (Intermediate III), which retains its interactions with M1 and His178. The reaction pathway of prolidase is a fairly complicated process with many components involved. After a proton is removed from the bridge between the two Mn2+ ions, the GlyPro substrate causes a conformational change as it binds to the active site. This GlyPro is held in place by hydrogen bonds formed by multiple amino acids in this structure. The Gly-N atom of the GlyPro substrate and the Gly-O atom of the peptide bond each interact with the Mn2+ ions, which are stabilized by additional amino acids, leading to polarization.

The first reference to Israel in non-biblical sources is found in the Merneptah Stele in c. 1209 BCE. The inscription is very brief and says: "Israel is laid waste and his seed is not". The inscription refers to a people, not an individual or nation state, who inhabit central Palestine or the highlands of Samaria. Some Egyptologists suggest that Israel appeared in earlier topographical reliefs, dating to the Eighteenth Dynasty or Nineteenth Dynasty (i.e. reign of Ramesses II) , but this reading remains controversial. In the Hebrew Bible, Israel first appears in Genesis 32:29, where an angel renames Jacob to Israel after Jacob fought with him. According to the folk etymology given in the text, Israel is derived from yisra, "to prevail over" or "to struggle with", and El, a Canaanite-Mesopotamian creator god that is tenuously identified with Yahweh. However, modern scholarship interprets El as the subject, "El rules/struggles", from sarar (שָׂרַר) 'to rule' (cognate with sar (שַׂר) 'ruler', Akkadian šarru 'ruler, king'), which is likely cognate with the similar root sara (שׂרה) "fought, strove, contended". Dr. Tzemah Yoreh clarifies that Israel is a combination of 'to strive with' (ש.ר.ה) and 'God' (אל) and that Jacob's name alternates between Jacob and Israel in the biblical narrative, even after his renaming, due to the authors having different opinions about Jacob's moral character. The gradual ethnonymic shift from "Israelites" to "Jews", regardless of their descent from Judah is made explicit in the Book of Esther (4th century BCE) of the Tanakh.
== Pharmacology == Infliximab is a purified, recombinant DNA-derived chimeric human-mouse IgG monoclonal antibody that consists of mouse heavy and light chain variable regions combined with human heavy and light chain constant regions. It has a serum half-life of 9.5 days and can be detected in serum 8 weeks after infusion treatment. Infliximab neutralizes the biological activity of TNF by binding with high affinity to the soluble (free floating in the blood) and transmembrane (located on the outer membranes of T cells and similar immune cells) forms of TNF, and inhibits or prevents the effective binding of TNF with its receptors. Infliximab and adalimumab (another TNF antagonist) are in the subclass of "anti-TNF antibodies" (they are in the form of naturally occurring antibodies), and are capable of neutralizing all forms (extracellular-, transmembrane-, and receptor-bound) of TNF. Etanercept, a third TNF antagonist, is in a different subclass (receptor-construct fusion protein), and, because of its modified form, cannot neutralize receptor-bound TNF. Additionally, the anti-TNF antibodies adalimumab and infliximab have the capability of lysing cells involved in the inflammatory process, whereas the receptor fusion protein apparently lacks this capability. Other monoclonal antibodies targeting TNF are golimumab, adalimumab, and certolizumab pegol. Etanercept also binds and inhibits the action of TNF, but is not a monoclonal antibody (it is instead a fusion of TNF-receptor and an antibody constant region).
=== Law Society review === In December 2005, the Law Society revealed that it has set up a committee, named Review Committee on Capital Punishment, to examine capital punishment in the country. The president of the society, Senior Counsel Philip Jeyaretnam, said that the main focus of the review was on issues regarding administering the death penalty such as whether it should be mandatory. A report of the review would be submitted to the Ministry of Law. On 6 November 2006, they were invited to give its views on proposed amendments to the Penal Code to the Ministry of Home Affairs. In their report, issued on 30 March 2007, they argued against the mandatory death penalty:
=== Gingivectomy by electrosurgery === Electrosurgery is defined as the intentional passage of high-frequency waveforms or currents through the tissues of the body to achieve a controllable surgical effect. It has been used in dentistry for more than 60 years. Electrosurgery may be used where a blood-free environment is required, providing that there is no bone coming into contact with the instrument and good oral hygiene is maintained. The technique is not widely used due to multiple factors including cost and lack of information available on the topic. It is essential to have the presence of a circuit in an electrosurgical unit, allowing current to flow. By changing the mode of activation of this current, electrosurgery may be used for the cutting or coagulation of soft tissues. The basic types of electrosurgical techniques are coagulation, desiccation, fulguration and electrosection (cutting). The majority of clinical operations are done by electrosection. There are two main types of electrosurgical units, monopolar and bipolar. In monopolar units a separate electrode is needed, usually in the form of an indifferent plate behind the patient's back. The current begins in the electrosurgical unit and flows to the oral site through a wire and then to the secondary electrode. Heat is produced on contact with the oral tissues and cutting results. Bipolar devices have two electrodes on their cutting tip and the current travels from one to the other, which removes the need for an indifferent plate. Bipolar devices make a wider cut.
=== Improving agricultural productivity === Iran's agricultural sector has faced declining productivity due to water shortages, outdated irrigation systems, and inefficient farming practices. Improving agricultural productivity is key to stabilizing food prices, and several steps can be taken. Modernizing Irrigation Techniques: Iran relies heavily on water-intensive crops like rice and wheat. The country also has one of the highest water withdrawal rates in the world. Transitioning to modern irrigation systems such as drip or precision farming can reduce water usage while increasing crop yields. Adopting New Agricultural Technologies: Investing in agricultural technology, including mechanization, better seeds (e.g., drought-resistant varieties), and soil fertility management, could significantly boost production efficiency. Government Support for Farmers: Providing education, subsidies, and financial assistance to small farmers can help them transition to more sustainable practices. Support programs targeting efficient use of resources would increase resilience against price shocks and reduce reliance on imports.

Ice formation concentrates solutes and can mechanically stress the peptide or damage the delivery device. Thawing afterwards may leave aggregates that are not visible to the eye. Refrigeration keeps the solution above its freezing point while slowing chemical degradation.
Size-exclusion chromatography is the most common technique, separating monomer from dimer and higher-order species by hydrodynamic volume. It is usually paired with ultraviolet detection and, for trace work, with multi-angle light scattering. Results depend on mobile-phase conditions, so method details matter when comparing datasets.
The detailed degradation profile under real-world temperature excursions and repeated handling is not fully described in the open literature. Relative contributions of photo-degradation versus thermal pathways are also incompletely mapped. Whether specific impurity species carry different biological activity is an open question rather than an established finding.
It is given either as a once-weekly subcutaneous injection or as an oral tablet taken once daily. The two forms use different absorption strategies, so they are not interchangeable on a milligram-for-milligram basis.
