Activation of the GLP-1 receptor couples to Gs signalling and raises intracellular cyclic AMP in pancreatic beta cells. The resulting insulin release depends on prevailing glucose concentrations, so the effect is greater when glucose is elevated. Receptor engagement also suppresses glucagon secretion and slows gastric emptying, which flattens post-meal glucose excursions. In the central nervous system, signalling in hypothalamic and brainstem regions is associated with reduced appetite and lower energy intake. Studies continue to examine effects on cardiac, renal and hepatic endpoints; whether those benefits are independent of weight change remains an open question.
Clinical development of this compound followed earlier short-acting GLP-1 analogues that required frequent injection. Once-weekly subcutaneous formulations entered use after 2017, and an oral formulation using a permeation enhancer later became available. The oral version pairs the peptide with sodium N-(8-[2-hydroxybenzoyl] amino) caprylate, a carrier that improves uptake across the gastric epithelium. Interest has expanded from glycaemic control into weight management and metabolic liver disease. Regulatory status and approved indications differ between countries, and the product remains subject to ongoing safety monitoring.
As a peptide, semaglutide is sensitive to conditions that break amide bonds or modify side chains. Deamidation of asparagine and glutamine residues, oxidation of methionine and tryptophan, and non-covalent aggregation are the main degradation routes described in published stability work. Rate depends strongly on pH, buffer species, ionic strength, temperature and exposure to light. Formulators therefore choose a defined solution pH and often add excipients such as phosphate buffer, propylene glycol and phenol, each of which plays a separate role in pH control, tonicity or preservation.
Storage guidance for the finished injectable product distinguishes the unused state from the in-use state. Before first use, pens are kept refrigerated between 2 and 8 degrees Celsius, protected from light, and never frozen, since freezing can disrupt the peptide or the device. After first use, label instructions in several markets permit storage at room temperature up to about 30 degrees Celsius for a limited number of days. Solid research-grade material is normally held at or below minus 20 degrees Celsius, often with desiccant, and allowed to equilibrate before opening.
Quantification and purity assessment rely on separation methods coupled to optical or mass detection. Reversed-phase high-performance liquid chromatography resolves the intact peptide from related impurities and is the standard assay technique. Size-exclusion chromatography measures aggregates, while ion-exchange chromatography separates charge variants produced by deamidation. Mass spectrometry confirms identity and detects mass shifts of a few daltons. In biological matrices, liquid chromatography with tandem mass spectrometry is often used because immunoassays can cross-react with endogenous GLP-1 or with circulating fragments.

Storage at minus 20 degrees Celsius or lower in a desiccated container preserves the peptide for extended periods, while working solutions are commonly held at two to eight degrees Celsius for short intervals. Light exposure and repeated freeze-thaw cycles accelerate degradation, so dividing material into single-use aliquots is generally recommended. Adsorption to glass and plastic surfaces can lower the measured concentration of dilute solutions, particularly below one milligram per millilitre. The degradation routes most often reported for GLP-1 analogues are deamidation, methionine oxidation, and backbone hydrolysis. Relative rates under specific conditions are frequently described only for individual formulations.
Reverse-phase high-performance liquid chromatography with ultraviolet detection near 214 or 280 nanometres is widely used to assess purity and to resolve related impurities. Liquid chromatography coupled to mass spectrometry confirms identity through the protonated molecular ion and fragment ions formed in tandem experiments. Capillary electrophoresis and peptide mapping after enzymatic digestion supply complementary information on charge variants and modification sites. Circular dichroism and nuclear magnetic resonance can report on secondary structure in solution. Absolute quantification usually depends on an external standard, and reported purity depends on the detection wavelength and integration parameters chosen.
Lyophilised material appears as a white to off-white cake or powder that is hygroscopic, and containers are usually equilibrated to room temperature before opening to limit condensation. Dissolution is performed in water, phosphate-buffered saline, or a mildly alkaline buffer, since solubility rises above neutral pH. Gentle inversion or low-speed mixing is preferred, because vigorous vortexing can promote surface denaturation and aggregation. Complete dissolution may require several minutes, and brief sonication is sometimes applied. Passing the solution through a 0.22 micrometre membrane removes particulates but does not by itself sterilise the liquid.
=== Tactics === Wenger was inspired by Borussia Mönchengladbach as a child, and was later influenced by Total Football, a playing style developed by Rinus Michels at Ajax in the 1970s. He recollected the team as having "perfect players everywhere and that was the sort of football I wanted to be playing myself". At Monaco, he employed a 4–4–2 formation, though he did trial 4–3–3, akin to Michels'. Wenger is an advocate of 4–4–2 as "no other formation is as efficient in covering space", but used it sparingly in his final seasons.

=== Reducing gastric reflux === The stomach produces gastric juice, a mixture consisting of gastric acid (mainly hydrochloric acid), lipase, and pepsin to enable food digestion. Constriction of the lower esophageal sphincter protects the esophageal mucosa by preventing reflux, the backflow of acid and gastric contents into the esophagus. The acute angle of His and the lower crura of the diaphragm also help this sphincteric action.

He obtained his doctorate in physics from the University of Amsterdam in June of 1950. Starting in 1947 he began work for Royal Dutch Shell as a researcher and it was there that he developed and published his article in 1956.
There are also a few thousand Druze immigrants from Lebanon in the United States of America, who have converted to Christianity. In the period of Egyptian rule in the Levant in the 1830s, many Druze converted to Christianity to avoid enlistment into the Egyptian army. The baptism of children in accordance with Christian custom was usual in large, well-known Lebanese Druze families, according to historian Aharon Layish there is also explicit evidence of Druzes in Lebanon under the Ottoman rule were posing Christians for practical reasons. The early Druze migrants from Levant to Venezuela tended to mix well with the local population, and some Druze converted to Catholicism. By one estimate made by Elisabe Granli from University of Oslo, around 1,920 Syrian Druze converted to Christianity; according to the same study, Christians with a Druze background (Druze converts to Christianity) still regard themselves as Druze, and claim that there is no contradiction between being Druze and being Christian. According to the Druze religious courts, between 1952 and 2009, around 10% of Israeli Druze who left the Druze faith converted to Christianity. According to Open Doors, there is a small but growing community of Druze converts to Christianity in Syria and Lebanon, with most converting to Evangelical Protestantism. These converts have established churches specifically for Christians of Druze background, primarily comprising women, girls, and young men who have abandoned the Druze religion they were raised in.
Jung was introduced to Freud's work by Bleuler whilst working at Burghölzli Hospital. Bleuler asked him to write a review of The Interpretation of Dreams (1899), which he read in 1900. In the early 1900s, psychology as a science was still in its early stages, but Jung became a qualified practitioner of Freud's new "psycho-analysis". Freud needed collaborators and pupils to validate and spread his ideas. Burghölzli was a renowned psychiatric clinic in Zurich, and Jung's research had already gained him international recognition. Jung sent Freud a copy of his Studies in Word Association in 1906. The same year, he published Diagnostic Association Studies, a copy of which he later sent to Freud, who had already purchased a copy. Preceded by a lively correspondence, Jung met Freud for the first time in Vienna on 3 March 1907.Jung recalled the discussion between himself and Freud as interminable and unceasing for 13 hours. Six months later, the then 50-year-old Freud sent a collection of his latest published essays to Jung in Zurich. This began an intense correspondence and collaboration that lasted six years. In 1908, Jung became an editor of the newly founded Yearbook for Psychoanalytical and Psychopathological Research. In the late summer of 1909, Jung sailed with Freud and Hungarian psychoanalyst Sándor Ferenczi to the United States. From 7–11 September, they took part in the twentieth-anniversary celebration of the founding of Clark University in Worcester, Massachusetts, the Vicennial Conference on Psychology and Pedagogy.
== Disease relevance == Homozygous or compound heterozygous mutations in 3-phosphoglycerate dehydrogenase cause Neu–Laxova syndrome and phosphoglycerate dehydrogenase deficiency. In addition significantly shortening lifespan, PHGDH deficiencies are known to cause congenital microcephaly, psychomotor retardation, and intractable seizures in both humans and rats, presumably due to the essential signaling within the nervous system that serine, glycine, and other downstream molecules are intimately involved with. Treatment typically involves oral supplementation of serine and glycine and has been shown most effective when started in utero via oral ingestion by the mother. Mutations that result in increased PHGDH activity are also associated with increased risk of oncogenesis, including certain breast cancers. This finding suggests that pathways providing an outlet for diverting carbon out of glycolysis may be beneficial for rapid cell growth. It has been reported that PHGDH can also catalyze the conversion of alpha-ketoglutarate to 2-Hydroxyglutaric acid in certain variants. Thus, a mutation in the enzyme is hypothesized to contribute to 2-Hydroxyglutaric aciduria in humans, although there is debate as to whether or not this catalysis is shared by human PHGDH. Research results suggest that PHGDH could serve as a blood biomarker of Alzheimer's disease.

The first documented use of electrical stimulation for pain relief dates back to 46 AD when Scribonius Largus of the ancient Roman Empire used the electric properties of torpedo fish to relieve headaches. Scientific studies of neuromodulation began in 1745, when German physician De Haen published "a number of cases of spasmodic, paralytic and other nervous affections cured by electricity". The first implementation of electrocutical apparatus in hospital medical treatment recorded in Middlesex Hospital of London in 1767. In 1780, Italian physicist and biologist Luigi Galvani investigated how electrical signals in nerves could control muscle movement, further evidence of the electrical nature of the neuronal activity.

In agriculture, Colombia is one of the five largest producers in the world of coffee, avocado and palm oil, and one of the 10 largest producers in the world of sugarcane, banana, pineapple and cocoa. The country also has considerable production of rice, potato and cassava. Although it is not the largest coffee producer in the world (Brazil claims that title), the country has been able to carry out, for decades, a global marketing campaign to add value to the country's product. Colombian palm oil production is one of the most sustainable on the planet, compared to the largest existing producers. Colombia is also among the 20 largest producers in the world of beef and chicken meat. Colombia is also the 2nd largest flower exporter in the world, after the Netherlands. Colombian agriculture emits 55% of Colombia's greenhouse gas emissions, mostly from deforestation, over-extensive cattle ranching, land grabbing, and illegal agriculture. Colombia is an important exporter of coal and petroleum – in 2020, more than 40% of the country's exports were based on these two products. In 2018 it was the 5th largest coal exporter in the world. In 2019, Colombia was the 20th largest petroleum producer in the world, with 791 thousand barrels/day, exporting a good part of its production – the country was the 19th largest oil exporter in the world in 2020.
Juan Calvete draws attention to this with explicitly when detailing the history of venomics. He declares that ''the last revolutions made in venomics research in the last decade (1989–1999) are the direct result of advancements made in proteomic-centered methods and the indirect result of more widely available and cost-effective forms of transcriptomics and bio-informatics analysis''. One of the first popular research topics of venomics was the pharmacological properties of the polypeptide toxins found in snake venom (Specifically, Elapidae and Hydrophidae) due to the neurotoxic properties and their ability to cause respiratory failure in animals. However, due to the lack of competent technology, less complex techniques (such dialysis to separate the venom), followed by simplistic chromatography and electrophoresis analysis, research was limited.
=== Oligonucleotide biotinylation === Oligonucleotides are readily biotinylated in the course of oligonucleotide synthesis by the phosphoramidite method using commercial biotin phosphoramidite. Upon the standard deprotection, the conjugates obtained can be purified using reverse-phase or anion-exchange HPLC

It belongs to the incretin mimetic class and acts as a long-acting glucagon-like peptide-1 receptor agonist. The class includes several peptides with different half-lives and routes of administration.
The side chain enables reversible binding to albumin in the bloodstream, which protects the peptide from rapid renal clearance. This extends the interval between administrations compared with unmodified GLP-1.
No. It shares much of the native sequence but carries substitutions and an added side chain. These changes increase stability against enzymatic breakdown.
Ice formation concentrates solutes and can mechanically stress the peptide or damage the delivery device. Thawing afterwards may leave aggregates that are not visible to the eye. Refrigeration keeps the solution above its freezing point while slowing chemical degradation.
