Degradation proceeds along several parallel routes. Deamidation of asparagine and glutamine residues generates charged variants that shift retention time in chromatographic analysis. Oxidation targets methionine and can be accelerated by trace metals or dissolved oxygen. Non-covalent aggregation produces dimers, oligomers, and larger species that are difficult to reverse. Isomerisation at aspartate residues is slower but measurable under thermal stress. The distribution among these pathways depends on pH, buffer composition, ionic strength, and the presence of excipients such as sugars or surfactants.
Reverse-phase high-performance liquid chromatography with ultraviolet detection near 214 nm is the standard purity method, reported as area percent. Mass spectrometry, usually with electrospray ionisation, confirms identity and reveals covalent modifications. Size-exclusion chromatography quantifies aggregates and fragments. Peptide mapping after enzymatic digestion localises changes to specific sequence regions. Circular dichroism and infrared spectroscopy report on secondary structure, while light scattering tracks particle formation in liquid formulations. No single technique captures every quality attribute.

Development began in the early 2010s with the goal of extending GLP-1 activity beyond the brief window achieved by native peptide infusion. The earliest approved formulation was a subcutaneous injection given once weekly. A later oral tablet pairs the peptide with an absorption enhancer, sodium N-(8-[2-hydroxybenzoyl] amino) caprylate, usually shortened to SNAC. That carrier lowers local pH and helps the peptide cross gastric tissue. Both routes deliver the same active molecule.
Semaglutide is a synthetic peptide analog of human glucagon-like peptide-1, a gut hormone released after meals. Its backbone retains the GLP-1 sequence but incorporates two substitutions that slow enzymatic breakdown by dipeptidyl peptidase-4. A short polyethylene glycol linker and a C18 fatty diacid are attached to the peptide chain, allowing the molecule to bind serum albumin and remain in circulation far longer than the native hormone. The result is a circulating half-life measured in days rather than the minutes typical of endogenous GLP-1.
Receptor activation occurs at GLP-1 receptors distributed across pancreatic islets, the hypothalamus, and the gastrointestinal tract. Binding triggers G protein signaling that raises cyclic AMP and enhances glucose-dependent insulin release. Because the effect depends on prevailing glucose levels, insulin secretion does not rise when blood sugar is already low. Signaling in the brain and gut also influences appetite and gastric emptying, which is why the compound appears in both metabolic and weight-related research literature.
The primary target is the GLP-1 receptor, a class B G protein-coupled receptor expressed on pancreatic beta cells, in the gut, and in several brain regions. Receptor activation raises intracellular cyclic AMP, which potentiates glucose-dependent insulin secretion and lowers glucagon release when blood glucose is elevated. Signalling in the hypothalamus and brainstem is associated with reduced appetite and slower gastric emptying. Because the insulinotropic effect depends on prevailing glucose levels, the hypoglycaemic risk of the peptide alone is described as low in most study settings. The relative contribution of peripheral and central actions remains an active research question.
Large randomised trials in adults with type 2 diabetes and in adults with obesity have reported reductions in body weight and improvements in several cardiovascular risk markers. One outcome trial found a lower incidence of major adverse cardiovascular events in participants with diabetes and established cardiovascular disease. Gastrointestinal effects such as nausea and vomiting are the most frequently reported adverse events and often diminish over time. Changes in lean body mass during weight loss are an area of ongoing investigation. Effects in adolescents and in pregnancy are less well characterised, and current labelling advises against use during pregnancy.
Certificate of analysis documents from suppliers typically report purity by chromatographic area, water content, and counter-ion identity. Independent verification is advisable because reported values can be generated under differing conditions. Impurity profiles matter for research use, where aggregates, deamidation products, and residual solvents may influence experimental results. Container, lot, and chain-of-custody records support traceability. Analytical results are method-dependent, so comparisons between laboratories require the same procedure and reference standards.
Lyophilized peptide material is typically stored at or below -20 °C, with -80 °C used for longer-term archives. Vials should remain sealed and desiccated because moisture promotes aggregation and hydrolysis. Repeated freeze-thaw cycles are avoided since they can alter peptide conformation and reduce recovery. Once reconstituted, solutions are generally kept at 2-8 °C and used within a defined window. Stability beyond those windows depends on buffer composition and concentration, and exact limits are product-specific rather than universal.
Activation of the GLP-1 receptor couples to Gs signalling and raises intracellular cyclic AMP in pancreatic beta cells. The resulting insulin release depends on prevailing glucose concentrations, so the effect is greater when glucose is elevated. Receptor engagement also suppresses glucagon secretion and slows gastric emptying, which flattens post-meal glucose excursions. In the central nervous system, signalling in hypothalamic and brainstem regions is associated with reduced appetite and lower energy intake. Studies continue to examine effects on cardiac, renal and hepatic endpoints; whether those benefits are independent of weight change remains an open question.
Clinical development of this compound followed earlier short-acting GLP-1 analogues that required frequent injection. Once-weekly subcutaneous formulations entered use after 2017, and an oral formulation using a permeation enhancer later became available. The oral version pairs the peptide with sodium N-(8-[2-hydroxybenzoyl] amino) caprylate, a carrier that improves uptake across the gastric epithelium. Interest has expanded from glycaemic control into weight management and metabolic liver disease. Regulatory status and approved indications differ between countries, and the product remains subject to ongoing safety monitoring.
The United States Congress established the Anti-Drug Abuse Act of 1988 to criminalize the distribution and possession of non-medical anabolic steroids. In 1999, WADA was formed to address the escalating use of substances in sports, particularly after the 1998 doping scandal in cycling.
The president of Serbia and Montenegro (Serbian: Предс(ј)едник Србије и Црне Горе, romanized: Preds(j)ednik Srbije i Crne Gore) was the head of state of Serbia and Montenegro. From its establishment in 1992 until 2003, when the country was reconstituted as a confederacy (state union) via constitutional reform, the head of state was known as the president of the Federal Republic of Yugoslavia (Serbian: Предс(ј)едник Савезне Републике Југославије, romanized: Preds(j)ednik Savezne Republike Jugoslavije). With the constitutional reforms of 2003 and the merging of the offices of head of government and head of state, the full title of the president was President of Serbia and Montenegro and Chairman of the Council of Ministers of Serbia and Montenegro (Serbian: Предс(ј)едник Србије и Црне Горе / Предс(ј)едник Сав(ј)ета министара Србије и Црне Горе). In 2006 the office was abolished as the state union was dissolved, with Serbia and Montenegro becoming independent countries.
2 O−2 + 2 H+ → O2 + H2O2 In biology this type of reaction is called a dismutation reaction. It involves both oxidation and reduction of superoxide ions. The superoxide dismutase (SOD) group of enzymes increase the rate of reaction to near the diffusion-limited rate. The key to the action of these enzymes is a metal ion with variable oxidation state that can act either as an oxidizing agent or as a reducing agent.
Smith had intended to have Dupont named Governor-General, but Queen Elizabeth II would not even consider this advice. With few exceptions, the international community backed Whitehall's assertion that Gibbs was the Queen's only legitimate representative, and hence the only lawful authority in Rhodesia. In September 1968, the Appellate Division of the High Court of Rhodesia ruled that Ian Smith's administration had become the de jure government of the country, not merely the de facto one. To support his decision, Chief Justice Sir Hugh Beadle used several statements made by Hugo Grotius, who maintained that there was no way that a nation could rightly claim to be governing a particular territory – if it was waging a war against that territory. Beadle argued that due to Britain's economic war against Rhodesia, she could not (at the same point) be described as governing Rhodesia. The ruling set the precedent that despite the UDI, the incumbent Smith government "could lawfully do anything its predecessors could lawfully have done". A Salisbury commission chaired by prominent lawyer W.R. Waley was appointed to study constitutional options open to the Rhodesian authorities as of April 1968, including on the topic of majority rule, but reopening negotiations with the British on a settlement was ruled out early on. The Waley Commission found that in practical as well as legal terms, "Europeans must surrender any belief in permanent European domination", pointing out that minority rule was not permanently sustainable.

=== Nosebleeds === Although oxymetazoline can be used for treatment of nosebleeds, it can also cause them under certain conditions, especially when nasal passages are dry. Oxymetazoline is a nasal decongestant that constricts blood vessels in the nasal mucosa, thereby reducing swelling and improving airflow. However, its use can lead to dryness and irritation of the nasal lining, which can increase the likelihood of nosebleeds.
==== European Union ==== European law stipulates that all member countries must allow the sale of irradiated dried aromatic herbs, spices and vegetable seasonings. However, these Directives allow Member States to maintain previous clearances food categories the EC's Scientific Committee on Food (SCF) had previously approved (the approval body is now the European Food Safety Authority). Presently, Belgium, Czech Republic, France, Italy, Netherlands, and Poland allow the sale of many different types of irradiated foods. Before individual items in an approved class can be added to the approved list, studies into the toxicology of each of such food and for each of the proposed dose ranges are requested. It also states that irradiation shall not be used "as a substitute for hygiene or health practices or good manufacturing or agricultural practice". These Directives only control food irradiation for food retail and their conditions and controls are not applicable to the irradiation of food for patients requiring sterile diets. In 2021 the most common food items irradiated were frog legs at 65.1%, poultry 20.6% and dried aromatic herbs, spices and vegetables seasoning. Due to the European Single Market, any food, even if irradiated, must be allowed to be marketed in any other member state even if a general ban of food irradiation prevails, under the condition that the food has been irradiated legally in the state of origin.

=== Mass spectrometry === Paper-spray ionization is being rapidly developed as an interface for micro paper-based analytical devices μPAD and mass spectrometry. The technique, first described by Graham Cooks group at Purdue, involves applying a voltage to a triangular sheet of wet paper near the inlet of a mass spectrometer. Although the exact mechanism is not well understood, two modes of operation can occur: a multicone spray at high flow rates, and a single cone spray that occurs when solvent has been depleted. This is part of a larger effort to combine complex microfluidic manipulations with mass spectral detection. Wax printing hydrophobic barriers is a common method for creating distinct flow channels within paper devices, and this has been extended to μPAD-MS to enhance ionization efficiency (by enabling focusing of the analyte stream) and enable reaction mixing by wax printing on the triangular paper surface. Chromatographic separations have also been demonstrated on μPADs prior to paper-spray detection. Initially, paper-spray ionization was applied for the detection of small molecules, such as pharmaceuticals and drugs of abuse. However, it has also been shown that paper-spray ionization can ionize large proteins while retaining non-covalent interactions.

Reverse-phase high-performance liquid chromatography with ultraviolet detection is the usual choice, with results reported as area percent. Complementary methods such as size-exclusion chromatography and mass spectrometry are needed because a single separation cannot resolve every impurity class. Purity figures are therefore method dependent and should always be read alongside the technique used.
Oxidation mainly affects methionine residues and is promoted by dissolved oxygen, trace transition metals, and prolonged exposure to light. Buffer choice and the presence of antioxidants in a formulation can alter the rate appreciably. Because the products differ in mass by only a few units, mass spectrometry is often required to detect them.
Published data on long-term ambient stability are limited, so the question remains open rather than settled. Short excursions during transport are common in practice, and many suppliers use insulated packaging with cold packs. Where stability data are absent, cold-chain handling with temperature logging is the safer approach.
