Clinical studies of semaglutide generally measure glycated hemoglobin, fasting plasma glucose, body weight, and composite cardiovascular endpoints. The SUSTAIN program enrolled adults with type 2 diabetes, while the STEP program focused on obesity without diabetes. Administration follows a stepwise escalation schedule designed to limit gastrointestinal effects during the first weeks. Reported outcomes include mean percentage weight change, the proportion of participants reaching defined weight-loss thresholds, and rates of nausea, vomiting, and diarrhea. Long-term data on durability after treatment stops are still limited and remain a topic of ongoing research.
Semaglutide is a synthetic peptide analog of human glucagon-like peptide-1, developed by Novo Nordisk and first approved in 2017 for type 2 diabetes. It belongs to the incretin mimetic class, a group of agents that reproduce the glucose-dependent actions of endogenous GLP-1. The molecule was engineered to resist degradation by dipeptidyl peptidase-4 and to bind serum albumin, extending its half-life from minutes to roughly one week. Approval for chronic weight management followed in 2021, based on large cardiovascular and obesity outcome trials.
Storage guidance for the finished injectable product distinguishes the unused state from the in-use state. Before first use, pens are kept refrigerated between 2 and 8 degrees Celsius, protected from light, and never frozen, since freezing can disrupt the peptide or the device. After first use, label instructions in several markets permit storage at room temperature up to about 30 degrees Celsius for a limited number of days. Solid research-grade material is normally held at or below minus 20 degrees Celsius, often with desiccant, and allowed to equilibrate before opening.
Quantification and purity assessment rely on separation methods coupled to optical or mass detection. Reversed-phase high-performance liquid chromatography resolves the intact peptide from related impurities and is the standard assay technique. Size-exclusion chromatography measures aggregates, while ion-exchange chromatography separates charge variants produced by deamidation. Mass spectrometry confirms identity and detects mass shifts of a few daltons. In biological matrices, liquid chromatography with tandem mass spectrometry is often used because immunoassays can cross-react with endogenous GLP-1 or with circulating fragments.
As a peptide, semaglutide is sensitive to conditions that break amide bonds or modify side chains. Deamidation of asparagine and glutamine residues, oxidation of methionine and tryptophan, and non-covalent aggregation are the main degradation routes described in published stability work. Rate depends strongly on pH, buffer species, ionic strength, temperature and exposure to light. Formulators therefore choose a defined solution pH and often add excipients such as phosphate buffer, propylene glycol and phenol, each of which plays a separate role in pH control, tonicity or preservation.

Native GLP-1 is degraded rapidly by dipeptidyl peptidase-4. Semaglutide resists this cleavage because alanine at position 8 is replaced by alpha-aminoisobutyric acid. A second substitution at position 34 introduces arginine, which further stabilizes the peptide. The most distinctive modification is a spacer and C18 fatty diacid attached at lysine 26, enabling strong albumin affinity. These three changes together produce a half-life measured in days rather than minutes, and the same structural logic underlies other long-acting analogs in this class.
Semaglutide is a synthetic peptide analog of glucagon-like peptide-1, a hormone released from intestinal L-cells after food intake. It contains 31 amino acids and differs from native GLP-1 through modifications that slow enzymatic breakdown. The peptide was developed to extend the short circulating half-life of endogenous GLP-1, which is measured in minutes. Researchers introduced the compound in the early 2010s. Two backbone changes and a fatty acid side chain define its structure, distinguishing it from earlier GLP-1 receptor agonists.
Identity and purity are assessed with reversed-phase high-performance liquid chromatography, which separates the peptide from related impurities by hydrophobicity. Mass spectrometry confirms molecular weight and detects truncation or modification products. Peptide mapping after enzymatic digestion verifies the amino acid sequence. Quantitation is often performed by LC-MS/MS or by immunoassay, and the two approaches can give different values because they measure different things. Method validation parameters such as accuracy, precision, and limit of quantitation are reported alongside results.
Certificate of analysis documents from suppliers typically report purity by chromatographic area, water content, and counter-ion identity. Independent verification is advisable because reported values can be generated under differing conditions. Impurity profiles matter for research use, where aggregates, deamidation products, and residual solvents may influence experimental results. Container, lot, and chain-of-custody records support traceability. Analytical results are method-dependent, so comparisons between laboratories require the same procedure and reference standards.
Semaglutide is a synthetic peptide analog of human glucagon-like peptide-1, a gut hormone released after meals. Its backbone retains the GLP-1 sequence but incorporates two substitutions that slow enzymatic breakdown by dipeptidyl peptidase-4. A short polyethylene glycol linker and a C18 fatty diacid are attached to the peptide chain, allowing the molecule to bind serum albumin and remain in circulation far longer than the native hormone. The result is a circulating half-life measured in days rather than the minutes typical of endogenous GLP-1.
Receptor activation occurs at GLP-1 receptors distributed across pancreatic islets, the hypothalamus, and the gastrointestinal tract. Binding triggers G protein signaling that raises cyclic AMP and enhances glucose-dependent insulin release. Because the effect depends on prevailing glucose levels, insulin secretion does not rise when blood sugar is already low. Signaling in the brain and gut also influences appetite and gastric emptying, which is why the compound appears in both metabolic and weight-related research literature.
== Side effects == As with other opioids, tolerance and physical and psychological dependence develop with repeated dihydrocodeine use. All opioids can impair the mental or physical abilities required for the performance of potentially hazardous tasks such as driving or operating machinery if taken in large doses. Itching and flushing and other effects of blood vessel dilation are also common side-effects, due to histamine release in response to the drug using one or more types of receptors in the CNS or other responses elsewhere in the body. First-generation antihistamines such as tripelennamine (Pyrabenzamine), clemastine (Tavist), hydroxyzine (Atarax), diphenhydramine (Benadryl), cyproheptadine (Periactin), brompheniramine (Dimetapp), chlorphenamine (Chlor-Trimeton), doxylamine (NyQuil) and phenyltoloxamine (Percogesic Original Formula) not only combat the histamine-driven side-effects, but are analgesic-sparing (potentiating) in various degrees. The antihistamine promethazine (Phenergan) may also have a positive effect on hepatic metabolism of dihydrocodeine as it does with codeine. Higher doses of promethazine may interfere with most other opioids with the exception of the pethidine family (Demerol and the like) by this or other unknown mechanisms. As with all drugs, side-effects depend on the person taking the medication. They can range in severity from mild to extreme, from headaches to difficulty breathing. Constipation is the one side-effect of dihydrocodeine and almost all opioids which is near-universal.
== Approved drugs == Tissue plasminogen activator (also known as tPA, t-PA, rtPA, Activase, or Alteplase or Actilyse) is a drug that breaks down blood clots. It was first approved in 1996, yet this drug has no generic competition. US sales of the drug under the brand name Activase and a similar drug were approximately US$1.3 billion in 2021, while European sales under the brand name Actilyse were an additional 448 million Euro in 2019. Edaravone (radicut) was approved in Japan in 2001. It has an unknown mechanism of action, but is hypothesized to act through its antioxidant properties.
In January 2020, astronomers reported that silicon carbide grains from the Murchison meteorite had been determined to be presolar material. The oldest of these grains was found to be 3 ± 2 billion years older than the 4.54 billion years age of the Earth and Solar System, making it the oldest material found on Earth to date.

== Definition == Stimulant is an overarching term that covers many drugs including those that increase the activity of the central nervous system and the body, drugs that are pleasurable and invigorating, or drugs that have sympathomimetic effects. Sympathomimetic effects are those effects that mimic or copy the actions of the sympathetic nervous system. The sympathetic nervous system is a part of the nervous system that prepares the body for action, such as increasing the heart rate, blood pressure, and breathing rate. Stimulants can activate the same receptors as the natural chemicals released by the sympathetic nervous system (namely epinephrine and norepinephrine) and cause similar effects. Psychostimulant refers to a subset of substances within the stimulant drug class that act at the dopamine transporter (DAT), a protein that normally clears dopamine from the synaptic cleft. Amphetamine, methylphenidate, cocaine, and modafinil are examples of psychostimulants that promote arousal and affect task performance in a dose-dependent manner. Psychostimulants act at DAT by inhibiting dopamine reuptake or by initiating transporter-mediated dopamine efflux (i.e., reverse transport), thereby producing indirect dopamine agonist effects in the central nervous system. The DAT-mediated actions of psychostimulants contrast with other central nervous system stimulants that can overlap in behavioural effects, such as caffeine, a drug that promotes wakefulness primarily through antagonism of adenosine receptors.
=== Possibility of mirror-image life === The possibility of mirror-image life has been discussed since Louis Pasteur's 1860 work on molecular asymmetry. Advances in organic chemistry and synthetic biology may, in the future, lead to the possibility of fully synthesizing a living cell from small molecules, which could enable synthesizing mirror-image cells from mirrored versions (enantiomers) of life's building-block molecules. Some important proteins in the central dogma of molecular biology have been synthesized in mirror-image versions, including DNA polymerase in 2016 and RNA polymerase in 2022. Reconstructing regular lifeforms in mirror-image form, using the mirror-image (chiral) reflection of their cellular components, could be achieved by substituting left-handed amino acids with right-handed ones, in order to create mirror reflections of proteins, and likewise substituting right-handed with left-handed nucleic acids. Because the phospholipids of cell membranes are also chiral, American geneticist George Church proposed using an achiral fatty acid instead of mirror-image phospholipids for the membrane. Electromagnetism, the dominant interaction in chemistry, is unchanged under mirror-image transformation (P-symmetry). There is a small alteration of weak interactions under reflection, which can produce very small corrections that theoretically favor the natural enantiomers of amino acids and sugars, but it is unknown if this effect is large enough to affect the functionality of mirror-image biomolecules or explain homochirality in nature.

Surface-supplied diving equipment (SSDE) is the equipment required for surface-supplied diving. The essential aspect of surface-supplied diving is that breathing gas is supplied from the surface, either from a specialised diving compressor, high-pressure gas storage cylinders, or both. In commercial and military surface-supplied diving, a backup source of surface-supplied breathing gas should always be present in case the primary supply fails. The diver may also wear a bailout cylinder (emergency gas supply) which can provide self-contained breathing gas in an emergency. Thus, the surface-supplied diver is less likely to have an "out-of-air" emergency than a scuba diver using a single gas supply, as there are normally two alternative breathing gas sources available. Surface-supplied diving equipment usually includes communication capability with the surface, which improves the safety and efficiency of the working diver. The equipment needed for surface supplied diving can be broadly grouped as diving and support equipment, but the distinction is not always clear. Diving support equipment is equipment used to facilitate a diving operation. It is either not taken into the water during the dive, such as the gas panel and compressor, or is not integral to the actual diving, being there to make the dive easier or safer, such as a surface decompression chamber. Some equipment, like a diving stage, is not easily categorised as diving or support equipment, and may be considered as either.

On 15 September 2022, during a speech commemorating El Salvador's 201st anniversary of independence, Bukele announced that he would run for re-election in 2024. According to Bukele, "developed countries have re-election, and thanks to the new configuration of the democratic institution of our country, now El Salvador will too". Constitutional lawyers criticized his announcement, saying that presidential re-election violates "at least" four articles of the El Salvador constitution.
As an expansion pack for Half-Life, Opposing Force is a first-person shooter. The overall gameplay of Opposing Force does not significantly differ from that of Half-Life. Players navigate through levels, fight hostile non-player characters and solve a variety of puzzles to advance. The game continues Half-Life's methods of an unbroken narrative. The player sees everything through the first person perspective of the protagonist and remains in control of the player character for almost all of the game. Story events are conveyed through the use of scripted sequences rather than cut scenes. Progress through the world is continuous; although the game is divided up into chapters, the only significant pauses are when the game needs to load the next part of an environment. Opposing Force also features an extended multiplayer, incorporating the various new environments and weapons into the original deathmatch mode used in Half-Life. After release, a new capture the flag mode with additional levels, items and powerups, was created by Gearbox. For the most part the player battles through the single-player game alone, but is occasionally assisted by friendly non-player characters. Security guards and scientists will occasionally help the player in reaching new areas and convey relevant plot information. However, Opposing Force also features fellow U.S. Marines who will assist the player in combat to a far greater degree than security guards.
==== Precipitation amount effect ==== In contrast to temperature control at high latitudes, the isotopic composition of precipitation in the tropics is mainly influenced by rainfall amount (negative correlation). This "amount effect" is also observed for summer precipitation in the subtropics. Willi Dansgaard, who first proposed the term "amount effect", suggested several possible reasons for the correlation: (1) As cooling and condensation progress, the rainfall isotopic composition reflects an integrated isotopic depletion by the Rayleigh process; (2) A small amount of rainfall is more likely to be influenced by evaporation and exchange with surrounding moisture, which tend to make it more isotopically enriched. At low latitudes, the amount effect for δ18O is around −1.6‰/100mm precipitation increase at island stations, and −2.0‰/100mm at continental stations. It was also noted that the amount effect was most pronounced when comparing isotopic composition of monthly precipitation at different places in the tropics. The amount effect is also expected for HIC, but there are not as many calibration studies. Across southeast Asia, the δD sensitivity to monthly precipitation amount varies between −15 and −25‰/100mm depending on location. In temperate regions, the isotopic composition of precipitation is dominated by rainfall amount in summer, but more controlled by temperature in the winter. The amount effect may also be complicated by changes in regional moisture sources.
Carod left the government after acknowledging that the meeting with ETA had taken place, but affirming that he had not negotiated anything, least of all a truce restricted to Catalonia. However, a few days later ETA declared a truce "only for Catalonia with effect from January 1, 2004."

It is given either as a once-weekly subcutaneous injection or as an oral tablet taken once daily. The two forms use different absorption strategies, so they are not interchangeable on a milligram-for-milligram basis.
Structural modifications, including a fatty acid side chain and non-natural amino acid substitutions, slow enzymatic breakdown and promote albumin binding. These changes support once-weekly dosing rather than twice-daily administration.
The pathways involving insulin, glucagon, gastric emptying, and appetite signaling are well described. How much each pathway contributes to weight reduction in a given person is not fully established.
Ice formation concentrates solutes and can mechanically stress the peptide or damage the delivery device. Thawing afterwards may leave aggregates that are not visible to the eye. Refrigeration keeps the solution above its freezing point while slowing chemical degradation.
