Reverse-phase high-performance liquid chromatography with ultraviolet detection near 214 or 280 nanometres is widely used to assess purity and to resolve related impurities. Liquid chromatography coupled to mass spectrometry confirms identity through the protonated molecular ion and fragment ions formed in tandem experiments. Capillary electrophoresis and peptide mapping after enzymatic digestion supply complementary information on charge variants and modification sites. Circular dichroism and nuclear magnetic resonance can report on secondary structure in solution. Absolute quantification usually depends on an external standard, and reported purity depends on the detection wavelength and integration parameters chosen.
Lyophilised material appears as a white to off-white cake or powder that is hygroscopic, and containers are usually equilibrated to room temperature before opening to limit condensation. Dissolution is performed in water, phosphate-buffered saline, or a mildly alkaline buffer, since solubility rises above neutral pH. Gentle inversion or low-speed mixing is preferred, because vigorous vortexing can promote surface denaturation and aggregation. Complete dissolution may require several minutes, and brief sonication is sometimes applied. Passing the solution through a 0.22 micrometre membrane removes particulates but does not by itself sterilise the liquid.
Quality control for peptide material focuses on identity, purity, content and the profile of impurities. Common degradants include deamidated and oxidised forms, plus aggregates formed during storage or handling. Forced degradation studies under heat, light, acid and peroxide help define which conditions accelerate change and which analytical methods detect it. Limits for individual impurities are set by pharmacopoeial monographs or manufacturer specifications. How much a given impurity affects biological activity is often uncertain, and conclusions may depend on the assay used.
Solid peptide material is generally kept at reduced temperature to limit degradation. Short-term storage at 2 to 8 degrees Celsius is common, while longer archival storage at minus 20 degrees Celsius or below is typical for lyophilised powder. Vials should remain sealed and protected from light, because ultraviolet exposure can oxidise susceptible residues. Repeated freeze-thaw cycles are avoided, as they promote aggregation and loss of soluble material. Solutions are less stable than solids and are usually prepared close to the time of use.

Large randomised trials in adults with type 2 diabetes and in adults with obesity have reported reductions in body weight and improvements in several cardiovascular risk markers. One outcome trial found a lower incidence of major adverse cardiovascular events in participants with diabetes and established cardiovascular disease. Gastrointestinal effects such as nausea and vomiting are the most frequently reported adverse events and often diminish over time. Changes in lean body mass during weight loss are an area of ongoing investigation. Effects in adolescents and in pregnancy are less well characterised, and current labelling advises against use during pregnancy.
Semaglutide is a synthetic peptide analogue of glucagon-like peptide-1, a gut hormone released after nutrient intake. The molecule contains 31 amino acid residues and differs from the native sequence at several positions. A non-natural residue at position eight resists the enzyme that normally truncates the hormone, while a lysine-linked fatty diacid side chain promotes binding to serum albumin. These two modifications extend the circulating half-life from minutes to roughly one week. The peptide is produced by solid-phase synthesis followed by selective acylation, and its identity and purity are confirmed by spectrometric and chromatographic techniques.
The primary target is the GLP-1 receptor, a class B G protein-coupled receptor expressed on pancreatic beta cells, in the gut, and in several brain regions. Receptor activation raises intracellular cyclic AMP, which potentiates glucose-dependent insulin secretion and lowers glucagon release when blood glucose is elevated. Signalling in the hypothalamus and brainstem is associated with reduced appetite and slower gastric emptying. Because the insulinotropic effect depends on prevailing glucose levels, the hypoglycaemic risk of the peptide alone is described as low in most study settings. The relative contribution of peripheral and central actions remains an active research question.
Clinical studies of semaglutide generally measure glycated hemoglobin, fasting plasma glucose, body weight, and composite cardiovascular endpoints. The SUSTAIN program enrolled adults with type 2 diabetes, while the STEP program focused on obesity without diabetes. Administration follows a stepwise escalation schedule designed to limit gastrointestinal effects during the first weeks. Reported outcomes include mean percentage weight change, the proportion of participants reaching defined weight-loss thresholds, and rates of nausea, vomiting, and diarrhea. Long-term data on durability after treatment stops are still limited and remain a topic of ongoing research.
Semaglutide is a synthetic peptide analog of human glucagon-like peptide-1, developed by Novo Nordisk and first approved in 2017 for type 2 diabetes. It belongs to the incretin mimetic class, a group of agents that reproduce the glucose-dependent actions of endogenous GLP-1. The molecule was engineered to resist degradation by dipeptidyl peptidase-4 and to bind serum albumin, extending its half-life from minutes to roughly one week. Approval for chronic weight management followed in 2021, based on large cardiovascular and obesity outcome trials.
GLP-1 receptors are expressed on pancreatic beta cells, in the gut, and in several brain regions. Receptor activation raises cyclic AMP, enhances glucose-dependent insulin secretion, and suppresses glucagon release when blood glucose is high. Effects on gastric emptying and on hypothalamic appetite circuits reduce energy intake. Because insulin release remains glucose-dependent, the risk of hypoglycemia is low when the drug is used alone. The precise contribution of each pathway to body weight change in humans remains an area of active investigation.
Receptor activation follows the canonical Gs pathway: binding increases intracellular cyclic AMP, which promotes protein kinase A activity. In pancreatic beta cells this amplifies glucose-dependent insulin release, so secretion rises when blood glucose is high and changes little when it is low. The same signalling suppresses glucagon release from alpha cells and slows gastric emptying. Receptors in the hypothalamus and brainstem are thought to contribute to reduced appetite and lower energy intake. Which of these effects dominates clinical outcomes remains an area of active study.
Semaglutide is a synthetic peptide analogue of glucagon-like peptide-1, a gut hormone released by intestinal L cells after food intake. The natural hormone acts on pancreatic and central receptors but is degraded within minutes by dipeptidyl peptidase-4 and other peptidases. Semaglutide belongs to the class of long-acting GLP-1 receptor agonists, a group distinguished by structural changes that slow breakdown and extend circulation time. Its development followed earlier short-acting analogues and reflects a general strategy in peptide drug design: preserve receptor activity while blocking proteolytic clearance.
Three structural changes define the molecule. At position 8 an alpha-aminoisobutyric acid residue replaces alanine, which blocks dipeptidyl peptidase-4 cleavage. At position 34 arginine replaces lysine, and at position 26 a lysine carries a C18 fatty diacid attached through a short linker. The fatty chain binds serum albumin, and this albumin association reduces renal filtration and enzymatic attack. The unchanged backbone retains the receptor contacts that produce signalling. The free base has the formula C187H291N45O59 and a molecular weight near 4114 daltons.
Development began in the early 2010s with the goal of extending GLP-1 activity beyond the brief window achieved by native peptide infusion. The earliest approved formulation was a subcutaneous injection given once weekly. A later oral tablet pairs the peptide with an absorption enhancer, sodium N-(8-[2-hydroxybenzoyl] amino) caprylate, usually shortened to SNAC. That carrier lowers local pH and helps the peptide cross gastric tissue. Both routes deliver the same active molecule.
Semaglutide is a synthetic peptide analog of human glucagon-like peptide-1, a gut hormone released after meals. Its backbone retains the GLP-1 sequence but incorporates two substitutions that slow enzymatic breakdown by dipeptidyl peptidase-4. A short polyethylene glycol linker and a C18 fatty diacid are attached to the peptide chain, allowing the molecule to bind serum albumin and remain in circulation far longer than the native hormone. The result is a circulating half-life measured in days rather than the minutes typical of endogenous GLP-1.
Receptor activation occurs at GLP-1 receptors distributed across pancreatic islets, the hypothalamus, and the gastrointestinal tract. Binding triggers G protein signaling that raises cyclic AMP and enhances glucose-dependent insulin release. Because the effect depends on prevailing glucose levels, insulin secretion does not rise when blood sugar is already low. Signaling in the brain and gut also influences appetite and gastric emptying, which is why the compound appears in both metabolic and weight-related research literature.
=== Acid hydrolysis === Acid hydrolysates are produced from various edible protein sources, with soy, corn, wheat, and casein being the most common. For the production of aHVP, the proteins are hydrolyzed by cooking with a diluted (15–20%) hydrochloric acid, at a temperature between 90 and 120 °C for up to 8 hours. After cooling, the hydrolysate is neutralized with either sodium carbonate or sodium hydroxide to a pH of 5 to 6. During hydrolysis, extraneous polymeric material known as humin, which forms from the interaction of carbohydrate and protein fragments, is generated and subsequently removed by filtration and then further refined. The source of the raw material, concentration of the acid, the temperature of the reaction, the time of the reaction, and other factors can all affect the organoleptic properties of the final product. Activated carbon treatment can be employed to remove both flavor and color components, to the required specification. Following a final filtration, the aHVP may, depending upon the application, be fortified with additional flavoring components. Thereafter, the product can be stored as a liquid at 30–40% dry matter, or alternatively it may be spray dried or vacuum dried and further used as a food ingredient. One hundred pounds (45 kg) of material containing 60% protein will yield 100 pounds of aHVP, which contains approximately 40 pounds (18 kg) of salt. This salt gain occurs during the neutralization step.
== Applications == PFBA has a variety of niche applications in analytical and synthetic chemistry. It is an ion pair reagent for reverse-phase high-performance liquid chromatography. It is used in the sequencing, synthesis, and solubilizing of proteins and peptides. Esters derived from PFBA readily undergo condensation, owing to their electrophilicity. Specialized ligands for metal ions are generated capitalizing on this property, such as Eufod. PFBA was formerly used for manufacturing photographic film. The 3M Company was a major manufacturer of PFBA and products containing PFBA but production was phased out in 1998.
==== 2011 Veracruz massacres ==== In spring 2011, the CJNG declared war on all other Mexican cartels and stated its intention to take control of the city of Guadalajara. However, by midsummer, the group appeared to have been reunited with its former partners in the Sinaloa Cartel. In addition to maintaining its anti-Zetas alliance with the Gulf Cartel, the Sinaloa Cartel in 2011 affiliated itself with the Knights Templar in Michoacán. To counter Los Zetas in the state of Jalisco, the Sinaloa Cartel affiliated itself with the CJNG. On 20 September 2011, two trucks containing 35 dead bodies were found at an underpass near a shopping mall in Boca del Río, Veracruz. All of the corpses were alleged to be members of Los Zetas, but it was later proven that only six of them had been involved in minor crime incidents, and none of them were involved with organized crime. Some of the victims had their hands tied and showed signs of having been tortured. According to El Universal, at around 17:00 an undetermined number of vehicles blocked a major avenue in Boca del Río. Once the traffic stopped, armed men abandoned two trucks in the middle of the highway. They opened the doors of the trucks and pulled out the thirty-five corpses, leaving a written message behind. Other gunmen pointed their weapons at the frightened drivers. The gunmen then fled the scene. Consequently, the stunned motorists began to grab their cellphones and post messages on Twitter warning other drivers to avoid the area. The message left behind stated the following:
==== Photografting ==== The modification of inert surfaces of polyolefins, polyesters, and polyamides by grafting functional vinyl monomers has been used to increase hydrophobicity, dye absorption, and polymer adhesion. This photografting method is generally used during continuous filament or thin film processing. On a bulk commercial scale, the grafting technique is referred to as photoinitiated lamination, where desired surfaces are joined by grafting a polymeric adhesion network between the two films. The low adhesion and absorption of polyolefins, polyesters, and polyamides is improved by UV-irradiation of an initiator and monomer transferred through the vapor phase to the substrate. Functionalization of porous surfaces have seen great success with high temperature photografting techniques. In microfluidic chips, functionalizing channels allows directed flow to preserve lamellar behavior between and within junctions. The adverse turbulent flow in microfluidic applications can compound component failure modes due to the increased level of channel interdependency and network complexity. In addition, the imprinted design of microfluidic channels can be reproduced for photografting the corresponding channels with a high degree of accuracy.
==== Acute myocardial infarction ==== Beta blockers are indicated for the treatment of acute myocardial infarctions. During a myocardial infarction, systemic stress causes an increase in circulating catecholamines. This results an increase in heart rate and blood pressure, therefore increasing myocardial oxygen demand. Beta blockers competitively inhibit catecholamines acting on the β1-adrenergic receptors, thus reducing these detrimental effects and resulting in reduced myocardial oxygen consumption and demand. A 2019 Cochrane review compared beta blockers with placebo or no intervention, it found that beta blockers probably reduced the short-term risk of reinfarction and the long-term risk of all-cause mortality and cardiovascular mortality. The review identified that beta blockers likely had little to no impact on short-term all-cause mortality and cardiovascular mortality.

The ultimate clinical candidate, D2E7, was created and manufactured at BASF Bioresearch Corporation and taken through most of the drug development process by BASF Knoll, then further development, manufacturing and marketing by Abbott Laboratories, after Abbott acquired the pharmaceutical arm of BASF Knoll. Since 2008, adalimumab had been approved by the FDA for the treatment of rheumatoid arthritis, psoriatic arthritis, ankylosing spondylitis, Crohn's disease, moderate to severe chronic psoriasis and juvenile idiopathic arthritis. Although only approved for ulcerative colitis from late 2012, by the FDA in the disease's management, it had been used for several years in cases that have not responded to conventional treatment at standard dosing for Crohn's disease. Adalimumab, sold under the brand name Humira, was approved for use in the United States in 2002. Adalimumab, sold under the brand names Humira and Trudexa, was approved for use in the European Union in September 2003.
=== Pentagon Christian worship services and "biblically sanctioned war" === In May 2025, Secretary Hegseth began holding monthly worship services during business hours at the Pentagon, and in February 2026 the Department of Defense began sending out invitations to defense contractors. Several stated that they feel that the services give Christians an advantage of "face time" with Pentagon officials that Jewish, Muslim or other non-Christians would not be given unless they attended the services, which they believe to be discriminatory. Service members have also reported complaints to the Military Religious Freedom Foundation about the services, and the Freedom From Religion Foundation has stated that the services show that Christian personnel are favored over their non-Christian peers in the Department of Defense. The Pentagon stated it did not track the names of persons who attended or did not attend the services. The worship services have included Doug Wilson, the founder of the Communion of Reformed Evangelical Churches, of which the secretary is a member. Wilson has stated that homosexuality should be a crime and that women should not be able to vote. In March 2026, soon after the start of the U.S. war against Iran, also known as Operation Epic Fury, it was reported that military leaders told their service members that the war was "part of God's divine plan" and that President Donald Trump was anointed by Jesus. One commander reportedly quoted the Book of Revelation and said the war will bring the second coming of Jesus Christ.
The Morgan State University Band Program consists of six ensembles: the marching band, symphonic band, symphonic winds, pep band, jazz ensemble, and jazz combo. Titled the Magnificent Marching Machine, the marching band has performed at Morgan State football games, NFL games, Presidential Inaugurations, World Series games and in regional and local television appearances. On November 28, 2019, the Magnificent Marching Machine performed during Macy's Thanksgiving Day Parade in New York City. They also performed at the 80th Anniversary and Commemoration of D-Day in Normandy, France on June 12, 2024.
Where a larger resolution indicates a better separation of peaks. This definition is used in a number of mass spectrometry texts. This use is also implied by the term "high-resolution mass spectrometry." A high value for resolution corresponding to good separation of peaks is similar to the convention used with chromatography separations, although the definitions are not the same. High resolution indicating better peak separation is also used in ion mobility spectrometry.
Other methods of refrigeration include the air cycle machine used in aircraft; the vortex tube used for spot cooling, when compressed air is available; and thermoacoustic refrigeration using sound waves in a pressurized gas to drive heat transfer and heat exchange; steam jet cooling popular in the early 1930s for air conditioning large buildings; thermoelastic cooling using a smart metal alloy stretching and relaxing. Many Stirling cycle heat engines can be run backwards to act as a refrigerator, and therefore these engines have a niche use in cryogenics. In addition, there are other types of cryocoolers such as Gifford-McMahon coolers, Joule-Thomson coolers, pulse-tube refrigerators and, for temperatures between 2 mK and 500 mK, dilution refrigerators.

Following extended discussions with Richwood Pharmaceuticals regarding the resolution of a large number of issues related to the company's numerous violations of FDA regulations, the FDA formally approved the first Obetrol labeling/sNDA revisions in 1996, including a name change to Adderall and a restoration of its status as an approved drug product. In 1997 Richwood Pharmaceuticals was acquired by Shire Pharmaceuticals in a $186 million transaction. Richwood Pharmaceuticals, which later merged with Shire, introduced the Adderall brand in 1996 as an instant-release tablet. In 2006, Shire agreed to sell rights to the Adderall name for the instant-release form of the medication to Duramed Pharmaceuticals. DuraMed Pharmaceuticals was acquired by Teva Pharmaceuticals in 2008 during their acquisition of Barr Pharmaceuticals, including Barr's Duramed division. The first generic version of Adderall IR was introduced to the market in 2002. Later on, Barr and Shire reached a settlement agreement permitting Barr to offer a generic form of the extended-release drug beginning in April 2009.
=== Thymus === The thymus gland is one of the first organs to degenerate in normal healthy individuals. Researchers from the University of Edinburgh have succeeded in regenerating a living organ that closely resembles a juvenile thymus in terms of structure and gene expression profile.
=== Kynurenine/tryptophan ratio === Changes in the ratio of kynurenine versus tryptophan are reported for many diseases like arthritis, HIV/AIDS, neuropsychiatric disorders, cancer and inflammations. The kynurenin/tryptophan is also an indicator for the activity of indoleamine 2,3-dioxygenase (IDO).

=== Liquid chromatography EI-MS === Two recent approaches for coupling capillary scale liquid chromatography-electron ionization mass spectrometry (LC-EI-MS) can be incorporated for the analysis of various samples. These are capillary-scale EI-based LC/MS interface and direct-EI interface. In the capillary EI the nebulizer has been optimized for linearity and sensitivity. The direct-EI interface is a miniaturized interface for nano- and micro-HPLC in which the interfacing process takes place in a suitably modified ion source. Higher sensitivity, linearity, and reproducibility can be obtained because the elution from the column is completely transferred into the ion source. Using these two interfaces electron ionization can be successfully incorporated for the analysis of small and medium-sized molecules with various polarities. The most common applications for these interfaces in LC-MS are environmental applications such as gradient separations of the pesticides, carbaryl, propanil, and chlorpropham using a reversed phase, and pharmaceutical applications such as separation of four anti-inflammatory drugs, diphenyldramine, amitriptyline, naproxen, and ibuprofen. Another method to categorize the applications of electron ionization is based on the separation technique which is used in mass spectroscopy. According to this category most of the time applications can be found in time of flight (TOF) or orthogonal TOF mass spectrometry (OA-TOF MS), Fourier transform ion cyclotron resonance (FT-ICR MS) and quadrupole or ion trap mass spectrometry.

Filtering removes particulate matter that can block columns or scatter light. A 0.22 micrometre membrane is typical, and the filter material should be checked for peptide adsorption.
Peptides can bind to glass and plastic, so a fraction of the material leaves the solution. The effect is proportionally larger in dilute samples and can bias quantitative results.
Reverse-phase high-performance liquid chromatography is the most widely reported approach. Purity figures should always be quoted together with the wavelength, gradient, and integration parameters used.
Lower temperatures slow hydrolysis, oxidation and aggregation reactions that degrade the molecule over time. Lyophilised powder is more tolerant than solution, but both benefit from controlled conditions.
