If you have been reading about GLP-1 receptor and want a single page that covers the useful parts, this is it: definitions, context, how it is studied, and the questions that come up repeatedly.

The primary target is the GLP-1 receptor, a class B G protein-coupled receptor expressed on pancreatic beta cells, in the gut, and in several brain regions. Receptor activation raises intracellular cyclic AMP, which potentiates glucose-dependent insulin secretion and lowers glucagon release when blood glucose is elevated. Signalling in the hypothalamus and brainstem is associated with reduced appetite and slower gastric emptying. Because the insulinotropic effect depends on prevailing glucose levels, the hypoglycaemic risk of the peptide alone is described as low in most study settings. The relative contribution of peripheral and central actions remains an active research question.
Large randomised trials in adults with type 2 diabetes and in adults with obesity have reported reductions in body weight and improvements in several cardiovascular risk markers. One outcome trial found a lower incidence of major adverse cardiovascular events in participants with diabetes and established cardiovascular disease. Gastrointestinal effects such as nausea and vomiting are the most frequently reported adverse events and often diminish over time. Changes in lean body mass during weight loss are an area of ongoing investigation. Effects in adolescents and in pregnancy are less well characterised, and current labelling advises against use during pregnancy.
Semaglutide is a synthetic peptide analogue of glucagon-like peptide-1, a gut hormone released after nutrient intake. The molecule contains 31 amino acid residues and differs from the native sequence at several positions. A non-natural residue at position eight resists the enzyme that normally truncates the hormone, while a lysine-linked fatty diacid side chain promotes binding to serum albumin. These two modifications extend the circulating half-life from minutes to roughly one week. The peptide is produced by solid-phase synthesis followed by selective acylation, and its identity and purity are confirmed by spectrometric and chromatographic techniques.
Clinical development of this compound followed earlier short-acting GLP-1 analogues that required frequent injection. Once-weekly subcutaneous formulations entered use after 2017, and an oral formulation using a permeation enhancer later became available. The oral version pairs the peptide with sodium N-(8-[2-hydroxybenzoyl] amino) caprylate, a carrier that improves uptake across the gastric epithelium. Interest has expanded from glycaemic control into weight management and metabolic liver disease. Regulatory status and approved indications differ between countries, and the product remains subject to ongoing safety monitoring.
Semaglutide is a synthetic peptide that acts as an agonist at the glucagon-like peptide-1 receptor. It is a structural analogue of human GLP-1(7-37), modified to resist enzymatic degradation by dipeptidyl peptidase-4. The peptide backbone contains alpha-aminoisobutyric acid at position 8, a substitution that stabilises the helix and slows cleavage. A fatty diacid side chain attached through a linker at lysine 34 promotes binding to serum albumin, which extends the circulating half-life. These two modifications together allow less frequent administration than native GLP-1 requires.
Activation of the GLP-1 receptor couples to Gs signalling and raises intracellular cyclic AMP in pancreatic beta cells. The resulting insulin release depends on prevailing glucose concentrations, so the effect is greater when glucose is elevated. Receptor engagement also suppresses glucagon secretion and slows gastric emptying, which flattens post-meal glucose excursions. In the central nervous system, signalling in hypothalamic and brainstem regions is associated with reduced appetite and lower energy intake. Studies continue to examine effects on cardiac, renal and hepatic endpoints; whether those benefits are independent of weight change remains an open question.

Reported molecular weight is approximately 4113.6 daltons for the free base, and the peptide is supplied as a lyophilized powder or in buffered liquid form depending on the intended use. It is freely soluble in water when formulated with appropriate excipients, though the unconjugated peptide shows limited stability at neutral pH over long periods. Analytical characterization typically relies on reversed-phase high-performance liquid chromatography and mass spectrometry. Purity specifications for research-grade material commonly exceed ninety-five percent by area. Isotopic and impurity profiles differ between suppliers.
Two principal therapeutic variants exist under separate regulatory filings, one indicated for glycemic control in type 2 diabetes and one for chronic weight management. Both use the same active molecule; differences lie in formulation strength, titration schedule, and labeling. Regulatory agencies in the United States and European Union approved injectable forms in 2017 and 2018 respectively. An oral tablet formulation received approval later, using a carrier molecule to enhance absorption across the gastric epithelium. Labeling differs by jurisdiction and by indication.
Serum protein binding dominates the pharmacokinetic profile. The attached chain associates strongly with albumin, shielding the peptide from enzymatic attack and slowing filtration by the kidney. This interaction extends the circulation half-life to roughly one week in humans, which supports weekly administration intervals. An oral version pairs the peptide with an absorption enhancer that transiently alters gastric epithelium, permitting limited uptake; bioavailability by that route is substantially lower than by injection.
Semaglutide belongs to the glucagon-like peptide-1 receptor agonist class, a group of synthetic peptides that imitate an incretin hormone released by intestinal L cells after food intake. Native GLP-1 circulates for only a few minutes because dipeptidyl peptidase-4 cleaves it rapidly. The hormone acts on pancreatic islets, the gastrointestinal tract, and several brain regions. Because the natural peptide is short-lived, development work concentrated on analogues that keep receptor activity while resisting enzymatic breakdown and renal clearance.
The semaglutide sequence is a 31-residue analogue of human GLP-1, altered at three positions relative to the parent hormone. Aminoisobutyric acid replaces alanine at position 8, arginine replaces lysine at position 34, and a lipophilic diacid is attached to lysine 26 through a short linker. These features are reported consistently in the structural literature. The position 8 substitution blocks recognition by dipeptidyl peptidase-4, while the attached chain drives strong, reversible association with a carrier protein in blood.
Reverse-phase high-performance liquid chromatography with ultraviolet detection near 214 or 280 nanometres is widely used to assess purity and to resolve related impurities. Liquid chromatography coupled to mass spectrometry confirms identity through the protonated molecular ion and fragment ions formed in tandem experiments. Capillary electrophoresis and peptide mapping after enzymatic digestion supply complementary information on charge variants and modification sites. Circular dichroism and nuclear magnetic resonance can report on secondary structure in solution. Absolute quantification usually depends on an external standard, and reported purity depends on the detection wavelength and integration parameters chosen.
Lyophilised material appears as a white to off-white cake or powder that is hygroscopic, and containers are usually equilibrated to room temperature before opening to limit condensation. Dissolution is performed in water, phosphate-buffered saline, or a mildly alkaline buffer, since solubility rises above neutral pH. Gentle inversion or low-speed mixing is preferred, because vigorous vortexing can promote surface denaturation and aggregation. Complete dissolution may require several minutes, and brief sonication is sometimes applied. Passing the solution through a 0.22 micrometre membrane removes particulates but does not by itself sterilise the liquid.
Storage at minus 20 degrees Celsius or lower in a desiccated container preserves the peptide for extended periods, while working solutions are commonly held at two to eight degrees Celsius for short intervals. Light exposure and repeated freeze-thaw cycles accelerate degradation, so dividing material into single-use aliquots is generally recommended. Adsorption to glass and plastic surfaces can lower the measured concentration of dilute solutions, particularly below one milligram per millilitre. The degradation routes most often reported for GLP-1 analogues are deamidation, methionine oxidation, and backbone hydrolysis. Relative rates under specific conditions are frequently described only for individual formulations.
====== Master's degree and Graduate Certificate Programs in Pharmacy Leadership ====== In 2016, the college became the first pharmacy school in the US to offer an online MS degree program and graduate certificate in pharmacy leadership. These programs combine leadership and professional development in health care business and management courses. These are the first programs at the university to be a partnership between three difference colleges:
Surface bodies of water provide environments that dry out and rewet. Wet-dry cycles concentrate prebiotic compounds and enable condensation reactions to polymerise macromolecules. Moreover, lakes and ponds receive detrital input from weathering of continental apatite-containing rocks, the most common source of phosphates. The amount of exposed continental crust in the Hadean is unknown, but models of early ocean depths and rates of ocean island and continental crust growth make it plausible that there was exposed land. Another line of evidence for a surface start to life is the requirement for Ultraviolet radiation (UV) for organism function. UV is necessary for the formation of the U+C nucleotide base pair by partial hydrolysis and nucleobase loss. Simultaneously, UV can be harmful and sterilising to life, especially for simple early lifeforms with little ability to repair radiation damage. Radiation levels from a young Sun were likely greater, and, with no ozone layer, harmful shortwave UV rays would reach the surface of Earth. For life to begin, a shielded environment with influx from UV-exposed sources is necessary to both benefit and protect from UV. Shielding under ice, liquid water, mineral surfaces (e.g. clay) or regolith is possible in a range of surface water settings.
In the body, elastin is usually associated with other proteins in connective tissues. Elastic fiber in the body is a mixture of amorphous elastin and fibrous fibrillin. Both components are primarily made of smaller amino acids such as glycine, valine, alanine, and proline. The total elastin ranges from 58 to 75% of the weight of the dry defatted artery in normal canine arteries. Comparison between fresh and digested tissues shows that, at 35% strain, a minimum of 48% of the arterial load is carried by elastin, and a minimum of 43% of the change in stiffness of arterial tissue is due to the change in elastin stiffness.
Traditionally in composite science a strong interface between the matrix and filler is required to achieve favorable mechanical properties. If this is not the case, the phases tend to separate along the weak interface and makes for very poor mechanical properties. In a MCC composite however this is not the case, if the interaction between the filler and matrix is stronger than the filler-filler interaction the mechanical strength of the composite is noticeably decreased. Difficulties in natural fiber nanocomposites arise from dispersity and the tendency small fibers to aggregate in the matrix. Because of the high surface area to volume ratio the fibers have a tendency to aggregate, more so than in micro-scale composites. Additionally secondary processing of collagen sources to obtain sufficient purity collagen micro fibrils adds a degree of cost and challenge to creating a load bearing cellulose or other filler based nanocomposite.
=== Imaging === Cartilage does not absorb X-rays under normal in vivo conditions, but a dye can be injected into the synovial membrane that will cause the X-rays to be absorbed by the dye. The resulting void on the radiographic film between the bone and meniscus represents the cartilage. For in vitro X-ray scans, the outer soft tissue is most likely removed, so the cartilage and air boundary are enough to contrast the presence of cartilage due to the refraction of the X-ray.

Thus, instead of letting the wave propagate through the fuel, the fuel itself is moved through a largely stationary burn wave. This is contrary to many media reports, which have popularized the concept as a candle-like reactor with a burn region that moves down a stick of fuel. By replacing a static core configuration with an actively managed "standing wave" or "soliton" core, TerraPower's design avoids the problem of cooling a highly variable burn region. Under this scenario, the reconfiguration of fuel rods is accomplished remotely by robotic devices; the containment vessel remains closed during the procedure, and there is no associated downtime. In a separate fast-reactor project, the NRC approved a construction permit for TerraPower's Natrium project at Kemmerer, Wyoming, in March 2026. The sodium-cooled fast reactor still required a separate operating licence before it could operate.
==== V-RG ==== RABORAL V-RG (Boehringer Ingelheim, Duluth, GA, USA) is a vaccinia virus (originally a smallpox vaccine) modified to express rabies glycoprotein. V-RG has been proven safe in over 60 animal species including cats and dogs. It is only approved for wildlife and sold to federal agencies. V-RG has been the only ORV licensed in the US for rabies virus management since 1997. V-RG baits consist of a small packet containing the oral vaccine which is then either coated in a fishmeal paste or encased in a fishmeal-polymer block.
=== Death and legacy === Several retrospective sources state that Liu was greatly saddened by Zhou Enlai's death in January 1976 and, according to Guangming Daily, even had himself carried on a stretcher to the Beijing Hospital to bid farewell to Zhou's remains. Having been diagnosed with cancer in 1975, Liu's own health deteriorated shortly after, and he died of his illness in Beijing on June 24, 1976. He was 81. His memorial service was held at the Babaoshan Revolutionary Cemetery and was presided by Vice Premier and general Chen Xilian. The June 30, 1976 issue of the People's Daily reported that Vice Chairperson Li Suwen delivered Liu's eulogy. According to the eulogy, Liu Wenhui supported Chairman Mao, the Cultural Revolution, the movement to criticize Lin Biao and Confucius, and finally the ongoing campaign against Deng Xiaoping. He had also called for the liberation of Taiwan from Kuomintang rule. Present at his funeral were several politicians including Wu De, Li Jingquan, Ngapoi Ngawang Jigme, and Zhou Jianren, among others. Marshals Zhu De, Ye Jianying, and Liu Bocheng sent wreaths to the funeral, as well as Hua Guofeng and Song Qingling.
=== American Cancer Society === Inhorn's ongoing interest in cervical cancer was the reason that he joined the American Cancer Society, Wisconsin Division, in 1967. He was Chair of the State Cancer Prevention Study II, and in 1986, he headed a demonstration project with a number of Wisconsin hospitals to determine the acceptance of offering low-cost mammography as a screening test for breast cancer. Inhorn served as President of the Wisconsin Division from 1970 to 1972, continuing on the board until 2010. He also served on the national American Cancer Society Board from 1982 to 1990.
== A scientific home in Pittsburgh == In a few short years, the Dean of the School of Medicine, himself a professor of Biochemistry, invited Hofmann to become Chairman of the Department. From the moment he took the position of Chairman of Biochemistry, it became clear that although he would always be a son of Switzerland, the United States offered him career opportunities he could never hope for in a small country like Switzerland. The US was to become his permanent home. The burgeoning field of peptide chemistry became his scientific focus and, in his own words, he fell in love with a molecule that was known to stimulate the adrenal cortex to produce the very steroids that had so fascinated him in Reichstein's laboratory. That molecule, not yet isolated, was ACTH and the love affair was lifelong. Despite detours into other areas, he kept returning to ACTH. In the last years before his death, he was developing methods to isolate the ACTH receptor.

== Gene regulation == MMP3 itself can enter in nuclei of cells and regulate target gene such as CTGF/CCN2 gene. Expression of MMP3 is primarily regulated at the level of transcription, where the promoter of the gene responds to various stimuli, including growth factors, cytokines, tumor promoters, and oncogene products. A polymorphism in the promoter of the MMP3 gene was first reported in 1995. The polymorphism is caused by a variation in the number of adenosines located at position -1171 relative to the transcription start site, resulting in one allele having five adenosines (5A) and the other allele having six adenosines (6A). In vitro promoter functional analyses showed that the 5A allele had greater promoter activities as compared with the 6A allele. It has been shown in different studies that individuals carrying the 5A allele have increased susceptibility to diseases attributed to increased MMP expression, such as acute myocardial infarction and abdominal aortic aneurysm. On the other hand, the 6A allele has been found to be associated with diseases characterized by insufficient MMP-3 expression due to a lower promoter activity of the 6A allele, such as progressive coronary atherosclerosis. The -1171 5A/6A variant has also been associated with congenital anomalies such as cleft lip and palate, where individuals with cleft lip/palate presented significantly more 6A/6A genotypes than controls.

== Epidemiology == Ehlers–Danlos syndromes are estimated to occur in about one in 5,000 births worldwide. Initially, prevalence estimates ranged from one in 250,000 to 500,000 people, but these were soon found to be low, as medical professionals became more adept at diagnosis. EDS may be far more common than the currently accepted estimate due to the wide range of severities with which the disorder presents. The prevalence of the disorders differs dramatically. The most common is hypermobile EDS, followed by classical EDS. The others are very rare. For example, fewer than 10 infants and children with dermatosparaxis EDS have been described worldwide. Some types of EDS are more common in Ashkenazi Jews. For example, the chance of being a carrier for dermatosparaxis EDS is one in 2,000 in the general population but one in 248 among Ashkenazi Jews. Some recent studies have found that EDS is significantly more prevalent in people who identify as transgender or gender-diverse, or who suffer from gender dysphoria. One review, of a pediatric EDS clinic in the American Midwest between 2020 and 2022, found that 17% of patients identified as trans or gender-diverse, 89% of whom were assigned female at birth. By comparison, roughly 1–2% of adolescents identify as trans or gender-diverse in the US overall. In a 2020 study, among adults undergoing gender-affirming surgery, 2.6% had a diagnosis of EDS—130 times the highest reported prevalence of EDS in the general population.
There is currently no use for any isotope of berkelium outside basic scientific research. Berkelium-249 is a common target nuclide to prepare still heavier transuranium elements and superheavy elements, such as lawrencium, rutherfordium and bohrium. It is also useful as a source of the isotope californium-249, which is used for studies on the chemistry of californium in preference to the more radioactive californium-252 that is produced in neutron bombardment facilities such as the HFIR. A 22 milligram batch of berkelium-249 was prepared in a 250-day irradiation and then purified for 90 days at Oak Ridge in 2009. This target yielded the first 6 atoms of tennessine at the Joint Institute for Nuclear Research (JINR), Dubna, Russia, after bombarding it with calcium ions in the U400 cyclotron for 150 days. This synthesis was a culmination of the Russia-US collaboration between JINR and Lawrence Livermore National Laboratory on the synthesis of elements 113 to 118 which was initiated in 1989.
the attempt to put into practice the radical theories of völkisch antisemitism from 1933 onward together with the policy following the beginning of the Second World War of forcibly moving around millions of people. the Nazi policies of dividing the population into those of genetic "value" and "non-value" in terms of education, social policy, health policy and demographics with the theme of "selecting" those with "value" over those of "non-value". the policies of "racial hygiene" of sterilizing the "genetically unhealthy" which was followed up by the Action T4 program launched in January 1939 of killing all mentally and physically disabled Germans, which provided the prototype for the extermination of the Jews. The Action T4 program of killing the disabled marked the first time that an entire group had been selected for extermination based solely for their perceived genetic flaws. starting with the conquest of Poland, the "forced employment of millions of foreign workers meant that the völkisch hierarchy of Herrenmensch and Untermensch became a structural feature of daily life" which provided a context for genocide as it desensitized much of the German public to the sufferings of others. the "escalation of terror" following the conquest of Poland in September 1939 and then by the "war of extermination" launched against the Soviet Union with Operation Barbarossa in June 1941 with Hitler giving the Commissar Order, unleashing the Einsatzgruppen to exterminate Soviet Jews, and the orders to allow millions of Soviet POWs to stave to death.

Native GLP-1 is degraded within minutes by dipeptidyl peptidase-4 and neutral endopeptidases. Semaglutide carries a non-natural amino acid at position eight that blocks that cleavage, and a fatty diacid side chain that binds albumin. The result is a much longer duration of action than the native hormone.
The fatty diacid chain associates strongly with serum albumin, which keeps the peptide in circulation and shields it from rapid renal clearance. Bound peptide is released gradually, producing a sustained receptor signal. This mechanism also reduces the peak-to-trough variation seen with shorter-acting analogues.
The balance between peripheral receptor activation and signalling in the central nervous system is not fully resolved. The extent to which slowed gastric emptying accounts for reduced energy intake, compared with direct effects on appetite circuits, is debated. Long-term effects on lean mass and on tissues outside the gut and brain are still under study.
It belongs to the incretin mimetic class and acts as a long-acting glucagon-like peptide-1 receptor agonist. The class includes several peptides with different half-lives and routes of administration.
