Clinical development of this compound followed earlier short-acting GLP-1 analogues that required frequent injection. Once-weekly subcutaneous formulations entered use after 2017, and an oral formulation using a permeation enhancer later became available. The oral version pairs the peptide with sodium N-(8-[2-hydroxybenzoyl] amino) caprylate, a carrier that improves uptake across the gastric epithelium. Interest has expanded from glycaemic control into weight management and metabolic liver disease. Regulatory status and approved indications differ between countries, and the product remains subject to ongoing safety monitoring.
Semaglutide is a synthetic peptide that acts as an agonist at the glucagon-like peptide-1 receptor. It is a structural analogue of human GLP-1(7-37), modified to resist enzymatic degradation by dipeptidyl peptidase-4. The peptide backbone contains alpha-aminoisobutyric acid at position 8, a substitution that stabilises the helix and slows cleavage. A fatty diacid side chain attached through a linker at lysine 34 promotes binding to serum albumin, which extends the circulating half-life. These two modifications together allow less frequent administration than native GLP-1 requires.

Lyophilized semaglutide is typically stored at temperatures between minus 20 and minus 80 degrees Celsius for long-term preservation. Short-term storage at 2 to 8 degrees Celsius is common for working aliquots. Repeated freeze-thaw cycles can degrade the peptide and are usually avoided. The molecule is hygroscopic in its solid form, so containers should remain sealed with desiccant. Solutions are less stable than powders and are generally prepared fresh. Light exposure is limited because aromatic residues can undergo photo-oxidation.
Semaglutide dissolves readily in water and in aqueous buffers near neutral pH. Solubility decreases near the isoelectric point, where net charge is minimal. Common laboratory solvents include phosphate-buffered saline and dilute ammonium bicarbonate. Strongly acidic or basic conditions may accelerate hydrolysis. Working concentrations are usually prepared by diluting a concentrated stock. Vial surfaces can adsorb small amounts of peptide at low concentrations, so carrier proteins or low-binding tubes are sometimes used.
Identity and purity are usually assessed by reverse-phase high-performance liquid chromatography coupled to mass spectrometry. Retention time and observed mass are compared against a reference standard run under identical conditions. Impurity profiles reveal deamidation products, oxidized methionine variants, and truncated fragments that arise during synthesis or storage. Peptide mapping through enzymatic digestion confirms the primary sequence, while amino acid analysis offers an independent check on overall composition.
Stability studies examine how temperature, pH, and moisture influence degradation rates over time. In aqueous solution, hydrolysis and deamidation accelerate as pH moves away from mildly acidic conditions. Light exposure and residual metal ions can also trigger oxidation of susceptible residues. Accelerated aging at elevated temperature is used to estimate shelf life, though extrapolation to room temperature carries uncertainty because individual degradation pathways do not always scale predictably.
Lyophilized semaglutide powder is typically held at minus twenty degrees Celsius for long-term storage. At that temperature, solid-state degradation reactions proceed slowly and the peptide remains intact for extended periods. Repeated freeze-thaw cycles are best avoided because they promote aggregation and can shift the proportion of monomeric peptide present. Working aliquots are often prepared so that each portion is thawed only once, and desiccant is placed inside the vial to limit moisture uptake.
Lyophilised material appears as a white to off-white cake or powder that is hygroscopic, and containers are usually equilibrated to room temperature before opening to limit condensation. Dissolution is performed in water, phosphate-buffered saline, or a mildly alkaline buffer, since solubility rises above neutral pH. Gentle inversion or low-speed mixing is preferred, because vigorous vortexing can promote surface denaturation and aggregation. Complete dissolution may require several minutes, and brief sonication is sometimes applied. Passing the solution through a 0.22 micrometre membrane removes particulates but does not by itself sterilise the liquid.
Storage at minus 20 degrees Celsius or lower in a desiccated container preserves the peptide for extended periods, while working solutions are commonly held at two to eight degrees Celsius for short intervals. Light exposure and repeated freeze-thaw cycles accelerate degradation, so dividing material into single-use aliquots is generally recommended. Adsorption to glass and plastic surfaces can lower the measured concentration of dilute solutions, particularly below one milligram per millilitre. The degradation routes most often reported for GLP-1 analogues are deamidation, methionine oxidation, and backbone hydrolysis. Relative rates under specific conditions are frequently described only for individual formulations.
Reverse-phase high-performance liquid chromatography with ultraviolet detection near 214 or 280 nanometres is widely used to assess purity and to resolve related impurities. Liquid chromatography coupled to mass spectrometry confirms identity through the protonated molecular ion and fragment ions formed in tandem experiments. Capillary electrophoresis and peptide mapping after enzymatic digestion supply complementary information on charge variants and modification sites. Circular dichroism and nuclear magnetic resonance can report on secondary structure in solution. Absolute quantification usually depends on an external standard, and reported purity depends on the detection wavelength and integration parameters chosen.

In 2025, Rui Qiu and colleagues named and described a second species of Sinosauropteryx: S. lingyuanensis. The assigned holotype is IVPP V 12415, which comprises a near complete skeleton of a potential juvenile which also preserves traces of feather integument. The specimen was collected around 2010 from the Lower Cretaceous Yixian Formation of Dawangzhangzi (Lingyuan, Western Liaoning, China), and though it is of juvenile nature it represents the largest reported Sinosauropteryx individual. The species name, lingyuanensis, refers to Lingyuan which is the city where the holotype specimen was discovered.
Berkelium is a synthetic chemical element; it has symbol Bk and atomic number 97. It is a member of the actinide and transuranium element series. It is named after the city of Berkeley, California, the location of the Lawrence Berkeley National Laboratory (then the University of California Radiation Laboratory) where it was discovered in December 1949. Berkelium was the fifth transuranium element discovered after neptunium, plutonium, curium and americium. The major isotope of berkelium, 249Bk, is synthesized in minute quantities in dedicated high-flux nuclear reactors, mainly at the Oak Ridge National Laboratory in Tennessee, United States, and at the Research Institute of Atomic Reactors in Dimitrovgrad, Russia. The longest-lived and second-most important isotope, 247Bk, can be synthesized via irradiation of 244Cm with high-energy alpha particles. Just over one gram of berkelium has been produced in the United States since 1967. There is no practical application of berkelium outside scientific research which is mostly directed at the synthesis of heavier transuranium elements and superheavy elements. A 22-milligram batch of berkelium-249 was prepared during a 250-day irradiation period and then purified for a further 90 days at Oak Ridge in 2009. This sample was used to synthesize the new element tennessine for the first time in 2009 at the Joint Institute for Nuclear Research, Russia, after it was bombarded with calcium-48 ions for 150 days. This was the culmination of the Russia–US collaboration on the synthesis of the heaviest elements on the periodic table.
== Middle Ages == With the fall of the western Roman Empire in 476, Italy was taken over by the Germanic chieftain Odoacer and later became part of the Ostrogothic Kingdom in 489. The Ostrogothic kings ruled officially as Magistri Militum of the Byzantine Emperors. All government and administrative positions were held by Romans, while all primary laws were legislated by the Byzantine Emperor. During the sixth century, under the Ostrogoths' rule, Romans remained the center of government and cultural life. The Roman Cassiodorus, like Boethius and Symmachus, emerged as one of the most prominent men of his time. He was an administrator, politician, scholar and historian who was born in Scylletium (near Catanzaro). He spent most of his career trying to bridge the divides of East and West, Greek and Latin cultures, Romans and Goths, and official Christianity and Arian Christianity, which was the form of Christianity adopted by the Ostrogoths and which had earlier been banned. He established the Vivarium (monastery) in Scylletium. He oversaw the collation of three editions of the Bible in Latin. Seeing the value of collecting all the books of the Bible in one volume, he was the first to produce Latin Bibles in single volumes. The most well-known was the Codex Grandior which became the ancestor of all modern western Bibles. Cassiodorus was the heart of the administration of the Ostrogothic kingdom.

Macrophages can internalize antigens through receptor-mediated phagocytosis. Macrophages have a wide variety of pattern recognition receptors (PRRs) that can recognize microbe-associated molecular patterns (MAMPs) from pathogens. Many PRRs, such as toll-like receptors (TLRs), scavenger receptors (SRs), C-type lectin receptors, among others, recognize pathogens for phagocytosis. Macrophages can also recognize pathogens for phagocytosis indirectly through opsonins, which are molecules that attach to pathogens and mark them for phagocytosis. Opsonins can cause a stronger adhesion between the macrophage and pathogen during phagocytosis, hence opsonins tend to enhance macrophages' phagocytic activity. Both complement proteins and antibodies can bind to antigens and opsonize them. Macrophages have complement receptor 1 (CR1) and 3 (CR3) that recognize pathogen-bound complement proteins C3b and iC3b, respectively, as well as fragment crystallizable γ receptors (FcγRs) that recognize the fragment crystallizable (Fc) region of antigen-bound immunoglobulin G (IgG) antibodies. When phagocytosing and digesting pathogens, macrophages go through a respiratory burst where more oxygen is consumed to supply the energy required for producing reactive oxygen species (ROS) and other antimicrobial molecules that digest the consumed pathogens.
The second step in lipid metabolism is absorption of fats. Short chain fatty acids can be absorbed in the stomach, while most absorption of fats occurs only in the small intestines. Once the triglycerides are broken down into individual fatty acids and glycerols, along with cholesterol, they will aggregate into structures called micelles. Fatty acids and monoglycerides leave the micelles and diffuse across the membrane to enter the intestinal epithelial cells. In the cytosol of epithelial cells, fatty acids and monoglycerides are recombined back into triglycerides. In the cytosol of epithelial cells, triglycerides and cholesterol are packaged into bigger particles called chylomicrons which are amphipathic structures that transport digested lipids. Chylomicrons will travel through the bloodstream to enter adipose and other tissues in the body.
==== 2017–2018 service disruptions and track improvements ==== Since the early 2010s, Amtrak had planned to fix the deteriorating rails and infrastructure around Penn Station, but due to the prioritization of other projects, applied only minimal fixes. In early 2017, this culminated in numerous power outages, derailments, and delays due to track maintenance delays. There were frequent service disruptions to train schedules caused by the deterioration of its tracks and their supporting infrastructure, as well as in those of the East River and North River tunnels that respectively connect the station to Long Island and New Jersey. A string of early 2017 service disruptions started on March 23, 2017, when an Acela train derailed, causing delays for the day. On April 3, a NJ Transit train derailed at a known problem site, where repairs had been deferred. This caused four days of reduced service along the Northeast Corridor for both Amtrak and NJ Transit, because the incident damaged the switch that connects Tracks 1–8 to the North River tunnels. This closure caused a cascading failure, delaying Amtrak and Long Island Rail Road trains on the unaffected tracks. On April 14, a New Jersey Transit train became stuck in the North River tunnels, causing the station to grow crowded with waiting passengers. After an Amtrak police officer used a Taser on a man who was acting disruptively, rumors of gunshots sparked a stampede that injured 16 people. Following the stampede, U.S. Senator Chuck Schumer called on Amtrak to centralize law enforcement response.

==== US death penalty ==== In August 2018, Nebraska became the first American state to use fentanyl to execute a prisoner. Carey Dean Moore, then one of the longest-serving death row inmates in the United States, was executed at the Nebraska State Penitentiary. Moore received a lethal injection, administered as an intravenous series of four drugs that included fentanyl citrate, to inhibit breathing and render the subject unconscious. The other drugs included diazepam as a tranquilizer, cisatracurium besylate as a muscle relaxant, and potassium chloride to stop the heart. The use of fentanyl in execution caused concern among death penalty experts because it was part of a previously untested drug cocktail. The execution was also protested by anti-death penalty advocates at the prison during the execution and later at the Nebraska State Capitol.
O-GlcNAc has been implicated in influenza A virus (IAV)-induced cytokine storm. Specifically, O-GlcNAcylation of S430 on interferon regulatory factor-5 (IRF5) has been shown to promote its interaction with TNF receptor-associated factor 6 (TRAF6) in cellular and mouse models. TRAF6 mediates K63-linked ubiquitination of IRF5 which is necessary for IRF5 activity and subsequent cytokine production. Analysis of clinical samples showed that blood glucose levels were elevated in IAV-infected patients compared to healthy individuals. In IAV-infected patients, blood glucose levels positively correlated with IL-6 and IL-8 levels. O-GlcNAcylation of IRF5 was also relatively higher in peripheral blood mononuclear cells of IAV-infected patients.
== History == Bob Hoffman launched Hoffman's Hi-Proteen Fudge and Hoffman's Hi-Proteen Cookies in the early 1950s, and competitor Joe Weider launched Candy Food Bars with bodybuilder Dave Draper on the packaging in the late 1960s. Pillsbury Space Food Bars and the bars by Tiger's Milk both came to market in the 1960s. Though they had protein, they were marked as energy or nutrition bars. PowerBar, invented in California in 1986, was an early protein bar.

It belongs to the incretin mimetic class and acts as a long-acting glucagon-like peptide-1 receptor agonist. The class includes several peptides with different half-lives and routes of administration.
The side chain enables reversible binding to albumin in the bloodstream, which protects the peptide from rapid renal clearance. This extends the interval between administrations compared with unmodified GLP-1.
No. It shares much of the native sequence but carries substitutions and an added side chain. These changes increase stability against enzymatic breakdown.
