=== Supply shortage === In 2011, the Sanofi Pasteur plant flooded, causing problems with mold. The facility, located in Toronto, Ontario, Canada, produced BCG vaccine products made with substrain Connaught such as a tuberculosis vaccine and ImmuCYST, a BCG immunotherapeutic and bladder cancer drug. By April 2012 the FDA had found dozens of documented problems with sterility at the plant including mold, nesting birds and rusted electrical conduits. The resulting closure of the plant for over two years caused shortages of bladder cancer and tuberculosis vaccines. On 29 October 2014 Health Canada gave the permission for Sanofi to resume production of BCG. A 2018 analysis of the global supply concluded that the supplies are adequate to meet forecast BCG vaccine demand, but that risks of shortages remain, mainly due to dependence of 75 percent of WHO pre-qualified supply on just two suppliers.

However, it shows differences from certain other antidepressants, like the tricyclic antidepressants, in animals. For example, it does not reverse the behavioral effects of the monoamine depleting agent reserpine and does not potentiate the effects of amphetamine or levodopa. Similarly to antipsychotics, trazodone reduces spontaneous motor activity, spontaneous and elicited aggressive behavior, and exploratory behavior, among other effects. In addition, trazodone diminishes amphetamine-induced locomotor hyperactivity, although it does not inhibit apomorphine- or amphetamine-induced stereotypy. On the other hand, unlike antipsychotics, trazodone does not produce catalepsy, although it can do so at sufficiently high doses. Activation of the serotonin 5-HT2A receptor enhances striatal dopaminergic neurotransmission, while stimulation of the serotonin 5-HT2C receptor inhibits striatal dopaminergic neurotransmission. Trazodone is both a serotonin 5-HT2A and 5-HT2C receptor antagonist, but has about 15-fold greater potency as an antagonist of the 5-HT2A receptor relative to the 5-HT2C receptor. === Physiological === Depending on the length of drug use, there is a chance that the immune system can be damaged. Most of these side-effects are dose-dependent, the most common being elevated blood pressure, especially in those with pre-existing hypertension. In addition to morphological changes of the heart which may have a permanent adverse effect on cardiovascular efficiency. AAS have been shown to alter fasting blood sugar and glucose tolerance tests. AAS such as testosterone also increase the risk of cardiovascular disease or coronary artery disease. Acne is fairly common among AAS users, mostly due to stimulation of the sebaceous glands by increased testosterone levels. Conversion of testosterone to DHT can accelerate the rate of premature baldness for males genetically predisposed, but testosterone itself can produce baldness in females. A number of severe side effects can occur if adolescents use AAS. For example, AAS may prematurely stop the lengthening of bones (premature epiphyseal fusion through increased levels of estrogen metabolites), resulting in stunted growth. Other effects include, but are not limited to, accelerated bone maturation, increased frequency and duration of erections, and premature sexual development.
